Significant effort has been invested in identifying pathways that regulate the activation of T cells. Among these pathways, the Salvador-Warts-Hippo (SWH) pathway stands out for its role in regulating cell proliferation and apoptosis, as well as its involvement in numerous functions of T cells, such as the formation of the immunological synapse and the memory of T lymphocytes. This pathway involves several families of protein kinases.
Beyond the core enzymes of this pathway, this article focuses on the thousand-and-one amino acid kinases (TAOKs) and SHP-1 (Figure 1). Modulation of SHP-1 abundance by TAOK3 thus serves as a rheostat for T cell receptor (TCR) signaling and determines the activation threshold of T lymphocytes.?
The results of the study presented in this article indicate that thousand-and-one-amino-acid kinase 3 (TAOK3) is a serine and threonine kinase that is essential for TCR signaling. Its absence results in reduced TCR signaling and increased interaction of the tyrosine phosphatase SHP-1 with the kinase LCK, a key downstream mediator of TCR signaling (Figure 1B).
To reach this conclusion, the scientific team of this study conducted precise molecular and biochemical analyses. Prior to these steps, it is crucial to isolate the enzyme of study in an optimal way in order to obtain the most reliable results.
