Accelerating cell line development by efficient and reliable single-cell isolation with DispenCell-S4


Cell line development is foundational to biotherapeutics, supporting the production of biologic drugs like monoclonal antibodies and AAVs for gene therapies. These therapies treat complex diseases with precision and effectiveness. For regulatory compliance, particularly with the FDA and IND submissions, cell lines must be monoclonal, with assurance of clonality to confirm each cell line's origin from a single cell. Additionally, cGMP readiness is essential to streamline the transition to CMC and scale-up production, ensuring biotherapeutics meet rigorous safety and quality standards for patients.

In this application note, we present a workflow for the isolation and culture of single cell clones using the DispenCell-S4 system and entirely disposable cGMP kits, showing that it provides fast and reliable isolation of single cell clones. This leads to highly efficient, reliable and traceable isolation of monoclonal cell lines, making the system particularly suitable for GMP environments and smooth IND filings.

1. What are the requirements of the medium used for cell sample preparation?

Most available culture media can be used. The medium conductivity range compatible with DispenCell-S4 is between 0.79 S/m and 1.71 S/m. If the medium used is known to contain insoluble particles, we recommend filtering it at 0.2 μm before using it to dilute the cells. To maximize the outgrowth, we recommend using a pre-warmed culture medium.

2. How should you proceed with cells that form aggregates?

Cell aggregates or clumps can cause clogging and reduce the efficiency of single cell isolation in all single cell dispensers including DispenCell. To mitigate this, we include cell strainers in our kits. However, additional aggregate removal protocols may be necessary if aggregates persist after filtration. Optimizing the suspension medium can help, such as using serum-free medium, Ca2?/Mg2?-free medium, or incorporating DNase, chelating agents (e.g., EDTA), or anti-clumping compounds into the process.
