In this edition of ACS Medicinal Chemistry Letters, Proteros scientists worked with Merck's R&D team to precisely express and elucidate the co-crystal structure of Merck's hIDO1 inhibitors. Crystal structure elucidation supported Merck's medicinal chemistry team in launching a structure-based drug design initiative allowing for low nM, potent inhibitors. Proteros Gallery Structures Highlight: CDK7
Cyclin-dependent kinase 7 (CDK7), Cyclin H, and RING-finger MAT1 form the heterotrimeric CDK-activating kinase (CAK) complex which is fundamental for transcription and cell-cycle control. Therapeutic intervention in cancer treatment has been exploited for CDK7 resulting in two selective inhibitors (THZ1 and its derivate SY-1365). These inhibitors can eliminate cancer cells that heavily rely on high levels of transcription such as triple-negative breast cancer. At Proteros we are currently offering CAK protein complex as cryo-EM Gallery Structure to support drug discovery programs of clients.
