In this edition of ChemMedChem, Proteros scientists worked with Boehringer Ingelheim's R&D team to elucidate the crystal structure of Linagliptin bound to DPP-4 and FAP-α. Crystal structure elucidation supported BI's medicinal chemistry team to understand the difference in substrate specificity and enzyme activity when only exchanging Ala657 with Asp663. Proteros Gallery Structures Highlight: hERG Channel
The hERG channel is a voltage-gated potassium channel involved in cardiac repolarization. Blocking of the hERG current prolongs ventricular?action potential duration?and delays?repolarization, potentially resulting in lethal arrhythmia. Off-target hERG inhibition by drugs is currently a critical issue in the pharmaceutical industry.
