Many life scientists are used to do ECL immuoassays. Either western or dot blots ore ELISA. ECL stands for enhanced chemiluminescence. Enhanced means that the signal is amplified, thus irregularities are amplified, too. Often life scientists have to fight with small airbubbles trapped, dust blocking smallest spots on the membrane, smooth sratches just from touching the membrane or similar minor irritations. Everything is amplified, the signal as well as the missing bits. That's why Particle Immunoassay utilizing nanosized gold is far more robust, simply because the signal does not require amplification, but is purely visible.
