Hello and welcome to our latest newsletter. In the Nordics we are longing for a bit more bright and warm days. As the days become longer, we certainly feel energized by the sun but also by meeting and talking with you. Last week Annette participated in a small exhibition in Link?ping and the coming week Lene and Veijo will be in Finland. In fact Iceland also had a visit last week by Katharina, so we are all filled with new experiences, which we would like to share with you. The focus this month will be oncology and an invite for a webinar.

Last month we shared our newest reagent kits- a lot was within the oncology area. Among those was AKT1 Phospho S129, CDS25C Phospho S216 and total, Cyclin A2 total, Cyclin B1 Phospho S126 and total, IKZF1 total, IKZF3 total.

The cell cycle is a series of events that a cell passes through to grow and divide. The cycle is split into distinct phases and depends on checkpoints to replicate and divide correctly.

The checkpoints rely on CDKs, cyclin-dependent kinases and Cycs, cyclins to slow or stop progression through the cell cycle if defects are detected. Critical checkpoints occur at different phases of the cell cycle: before DNA replication (GS/1), after DNA replication (G2/M), and during mitosis (spindle assembly checkpoint -SAC).

Dysregulation of the cell cycle and checkpoints can lead to diseases, such as cancer – where cells have sustained proliferation.

G1/S checkpoint - checks for DNA damage, controlling entry from G1 phase into S phase ( where DNA is replicated)

G2/M checkpoint - checks for DNA damage, controlling entry into M phase (where chromosomes are segregated)

SAC checkpoint – checks which chromosomes are correctly attached to the spindle, controlling entry into anaphase and commitment to cell division (cytokinesis)

Most cancer mutations occur before the S phase and can prevent cell cycle exit.

50-70% of breast cancers have Cyc D1 overexpression, promoting cell cycle progression.

Another interesting angle is how gut microbiome can act as an immune response modulator, when some patients remain unresponsive to treatment of immunotherapy. How can bacteria play a role in cancer progression, are the interactions localized in the gastrointestinal tract or do they occur in other tissues before malignancy. In relation to oncology we need to look into the following ? Apoptosis ? Cell cycle ? Cell viability ? DNA synthesis ? Cytotoxicity ? Cell migration ? Cell morphology ? Immunophenotyping ? Metabolism ? Wound healing ? Chemotaxis

So how do you count your cells - what about an image cytometer, that captures cellular images in brightfield and fluorescence for quantification, in terms of number, morphology, and fluorescence intensity. The image provides visual context to understand the quantitative results and enable concrete comparison between experimental conditions. The Celigo system can be utilized to monitor the efficacy of targeted cancer cell killing in a variety of complex assay formats. The Celigo can be implemented to characterize and screen a vast array of immunotherapy targets with ready- to-use signal-to-noise assays that will take you from imaging to quantitative results in less than ten minutes. Sensitive whole-well cell level analysis in brightfield and up to four color fluorescence for both adherent and suspension cells. Built-in gating function allows flow like population analysis directly in multiwell plates without the need to terminate cell culture and using only a small amount of sample. This enables ongoing monitoring of patient sample expansion and post-transformation proliferation as well as assessment of cell handling conditions such as cryopreservation and recovery protocols.
