The need for a multifunctional drug to treat TBI

Physical trauma to the head can seem straightforward, but injuries that affect the brain are never simple. Traumatic Brain Injury, TBI, is a global cause of affliction. To this day, no effective drug for treating traumatic brain injury has been approved by FDA, or by regulatory bodies within the EU.
The difficulty in developing a drug to treat TBI lies in the complexity of brain injuries. TBI is a heterogeneous disorder and progresses over time. The effects of a single point of injury can cascade in multiple ways (secondary brain damage) and damage ten times more nerve cells around the original area of injury. Although several inventive drugs have been researched to stop certain mechanisms of brain injury, such as Traxoprodil from Pfizer, or Dexanabinol from Pharmos, they have all failed to cure or prevent TBI and benefit the patients. In fact, more than 30 phase III clinical trials have failed only in the last few decades, because stopping a single cascade of biochemical events isn’t enough for patient recovery.

3 年Head injury and its outcome is different in every case since the damage to the tissue is different. Our personal physiology and health vary and the quality of the initial medical intervention is not uniform. The secondary damage to the tissue uninjured in the assault can continue for several weeks and months. The damage to our functions and the psychiatric effect varies with the location of the initial injury. In other word two cases alike.

3 年This is a welcomed approach. Having volunteered in a TBi rehabilitation centre for 7+ years, and being a survivor myself, I know first hand how the story doesn't end when one leaves the hospital.
