IMBAS-MS Discovers Organ-Specific HLA Peptide Patterns in Plasma

Immunopeptidomics, the study of peptides presented by human leukocyte antigen (HLA) proteins, has immense potential for biomarker discovery and understanding immune responses. However, current techniques are limited by complex workflows and low sensitivity, typically requiring large volumes of tissue or biological fluids. In this paper,? Maria Wahle et. al, introduce IMBAS-MS (Immunopeptidomics by Biotinylated Antibodies and Streptavidin), a highly sensitive, automated, and economical workflow for HLA peptide analysis from just 200 μl of plasma.

The key innovation of IMBAS-MS is the replacement of the traditional ProteinA/G-based antibody capture with a streptavidin-biotin system, simplifying the protocol and enabling generic use of biotinylated antibodies, in addition to multiplexing sample analysis using chemical labelling. Coupled with recent advancements in mass spectrometry, such as the Bruker timsTOF Ultra and data-independent acquisition (DIA) using predicted spectral libraries, IMBAS-MS quantifies over 5,000 HLA class I peptides in just 30 minutes, a substantial improvement over previous methods.

Through deep profiling of healthy donors, the authors demonstrate that the plasma immunopeptidome is remarkably stable over a year, exhibiting high reproducibility between individuals with overlapping HLA types. Intriguingly, immunopeptides originating from diverse tissues, except the brain, are proportionately represented, suggesting that the plasma immunopeptidome reflects system-wide changes in the human proteome.

This groundbreaking workflow paves the way for novel discoveries in immunology and precision oncology. IMBAS-MS could be used as a powerful tool for biomarker discovery, enabling minimally invasive monitoring of immune responses in various pathological conditions, from autoimmune disorders to cancer. With its enhanced sensitivity, throughput, and cost-effectiveness, IMBAS-MS could revolutionize the field of immunopeptidomics and become an invaluable asset in the pursuit of personalized medicine.
