To overcome these challenges, we established an automated, machine learning-enabled compound screening workflow with PDOs using the Gri3D? platform from SUN bioscience. The data revealed significant cytotoxic effects of certain compounds and demonstrated the potential for performing drug screening using robust and unbiased data analysis.

We’ve come a long way since the development of the first mouse intestinal organoids in 2009. However, organoid models still heavily rely on the use of solid extracellular matrix (ECM). This conventional culture method introduces a high level of heterogeneity in terms of size, shape, and distribution of organoids which complicates subsequent downstream readouts and image analyses.
In this application note, we share how you can overcome these challenges—and closely follow human rectal organoid development—using ready-to-use Gri3D? technology. This platform enables the homogeneous generation of a single microtissue in each microcavity in suspension-like conditions, without the need of a solid ECM.
