Here's a funny thing I've seen in the lab—someone do 100 cycles of PCR.
The person was trying to introduce a point mutation using inverse PCR. After 28 cycles, there was no PCR product on their agarose gel. So they put the PCR reaction back on for a further 72 cycles.?
They got a PCR product and when they transformed, prepped, and sequenced it, it had the desired mutation!?
Sometimes, the silly approaches work.
