Enzyme Services at Biosynth

Enzymes are versatile and essential components in biological life. Their involvement in such a wide range of biological processes such as digestion, metabolism, DNA replication and repair, cellular respiration and signal transductions. This makes them biomolecules of interest in many areas of scientific research. It is also the diversity, specificity, efficiency and reusability that make enzymes useful tools in fine chemistry, food and feed, pharma, cosmetics and textile chemistry industries. Enzymes can be used in the production of antibody-drug conjugates and many other applications, including:

  • Beta-lactamase enzymes can be used for environmental monitoring i.e. the growth of microbes in sterile facilities.
  • Nitrile hydratase has the potential to remove toxic nitriles from industrial waste water and the environment.
  • Lipases are useful in the synthesis of APIs and emollient esters for cosmetics.
  • Phospholipase D enzymes are used to synthesize critical raw materials for industrial, medical and cosmetic use.
  • Peroxidase enzymes are highly beneficial in immunoassay diagnostic tools.

This is just a tiny window into the significance and many applications of enzymes. Biosynth is dedicated not only to supplying tailored enzymes, but also to enhancing our customers' projects or industrial applications. Our enzyme team are leading experts in customized service solutions for enzymes. We guarantee to support you every step of the way, from the discovery and selection of your protein or enzyme right up to the final product formulation and manufacturing. Visit our enzyme webpage to discover our full enzyme offering, from catalog products to enzyme services and technologies.

Target Enzyme Identification

To begin our enzyme services timeline, Biosynth expertly designs suitable screening assays to identify novel enzymes for your bioconversion reactions. Using Frances H. Arnold’s “first law of random mutagenesis: You get what you screen for” Acc. Chem. Res. (1998), Biosynth can screen candidate enzymes from your own enzyme collections. Alternatively, we can perform general database or library screening. We are flexible to your needs. Enzyme libraries, proteins and microorganisms of your choice will then be expressed, isolated or cloned and screened.

Our customers can even carry out their own biocatalysis application tests using our reliable wide range of screening kits available from our enzyme product catalog.

Enzyme and Protein Engineering

Biosynth prides itself in developing its own proprietary in vivo recombination and other mutagenesis technologies to engineer a protein or enzyme for optimum performance. Our ethos is to assist customers in the life sciences industries in achieving remarkable industrial and research advances. We are therefore proud to share our engineering toolbox, equipped to provide customized proteins and enzymes with improved activity, stability and even substrate selectivity:

  • In silico analysis of protein structures and sequences.
  • Targeted site-directed and site-saturation mutagenesis.
  • In vivo recombination.
  • Gene shuffling.
  • Random mutagenesis.

Random, site-directed or site-saturation mutagenesis are our most popular technologies. However, for a more biodiverse and high-performing biocatalyst for pharma or industrial applications, our team are accomplished in gene shuffling and in vivo recombination technologies. The use of such technologies is fulfilling the growing demand for proteins and biopharmaceuticals to be produced with an improved yield and at a lower cost on an industrial scale. This allows for more options and effective treatments for diseases and disorders. Visit our enzyme technologies page to explore our protein and enzyme engineering capabilities.

High throughput screening for desired properties

Our enzyme specialists will work with you to ensure that the quality, activity, reaction kinetics and selectivity of the target enzyme are apt for your individual application. We perform in-depth analytical methods under our ISO9001 quality management system to characterize our proteins and enzymes under process-relevant conditions. The following analytical methods are available:

  • Protein analytics (PAGE, Western blot, Analytical gel filtration, HPLC-MS, etc.).
  • Differential scanning calorimetry.
  • High-resolution mass spectrometry.
  • Activity analysis with various substrates (HT assays, HPLC-MS, GC-MS, TLC).
  • Purity and contaminant analysis (HPLC-MS, Bioburden, TYMC, Heavy Metals, Endotoxin, TAMC, Sterility and Leachables).

Bioprocess development and production

Available microbial strains for use in Biosynth's bioprocess development.

In order to produce your specific protein, enzyme, recombinant protein or small molecule, Biosynth will craft reliable microbial fermentation methods and bioprocesses, selecting from a variety of microbial strains. We implement the following critical stages during our process development service:

Manufacture and Final Formulation

Our team will meticulously manufacture and deliver your product under agreed timelines to the highest quality using ISO9001 process controls. We also offer filling, labelling and packaging services for your final product.

Depending on our customer’s requests, our novel fermentation processes, downstream processing and tailored formulations can produce protein and enzymes from small to large scale. For example, our fermentation processes can be conducted up to the 1000L scale, and with our European partners, we can exceed this limit.

Although our production facility operates under non-GMP conditions, it is possible for us to transfer to GMP to meet our customer’s needs. Contact us to discuss how we can work with you to up-scale your challenging protein purification protocols and manufacture your target protein.

Final Formulation

The final formulation of your enzyme or protein is just as important as screening for and developing the most efficient bioprocesses for your biocatalytic solution. That is why we will work with you to choose the most appropriate final formation to suit your intended application. Whether your application is in fine chemistry, food and feed, pharma, cosmetics or textile chemistry industries, we can deliver your enzyme in a liquid, dry or immobilized formulation. We can even provide compatible buffers with additives for increased stability and activity, depending on your requirements.

Immobilizing your biocatalysts

We give our customers the option of immobilizing their biocatalysts, which offers clear benefits over free enzymes. Immobilized biocatalysts can be more easily removed from reaction mixtures by filtration or sedimentation. They have increased stability in challenging reaction environments, and finally, it enables biocatalysts to be recycled and reused. Overall, immobilization makes the production processes more cost-efficient.

Depending on your application, we can develop your immobilized biocatalysts using either:

  • Hydrophobic resins for adsorption of the respective enzyme (often used in reactions using organic solvents).
  • Chemically activated resins for covalent immobilization (used to prevent leaching of the enzyme in aqueous environments). ?

Access to speciality resins and particles from Mitsubishi Chemicals company, Resindion, Purolite or Evonik and in collaboration with these manufacturers allows us to tailor the immobilization method to your enzyme of choice and application. This allows us to offer our customers novel, tailored resin formulations and large-scale capacities for immobilization to ton-scale, thus increasing our flexibility to fulfil all industrial requirements. Find out more about the advantages of Biosynth's immobilized catalysts on our technologies page.
