Concentration-Response Testing by Automated Microscopy

Any therapeutic drug that alters the abundance, localization, morphology, distribution, or function of a protein, macromolecular structure or organelle within living cells can be assayed through automated fluorescence microscopy. One commonly employed approach is to evaluate drug effects at different concentrations, which is commonly called concentration-response testing or dose-response testing. The figure below demonstrates different expression levels of a protein in response to different concentrations of a drug, with higher expression levels of the protein after incubation with high drug concentrations. This result can be quantified and plotted as a concentration-response curve. We routinely conduct such assays in 384-well format, testing many compounds in parallel, which enables us to compare concentration-response curves of each compound and rank-order their potency.
