Insights on Stress and Age-Related Disease | A Customer Study Using a Nematode Animal Model

Nematodes may not have a lot to worry about, but they're at the forefront of research on proteotoxic stress in neurodegenerative disease! In 2022, Psomagen customers created an animal model in C. elegans to test the effects of a gate-destabilizing mutation on the 20S proteasome.

The resulting nematode population had enhanced proteasomal activity, including peptide, unstructured protein, and ubiquitin-dependent degradation activities. Subjects lived much longer than control populations, and were more resistant to oxidative and proteotoxic stress. Researchers used Psomagen's whole genome sequencing and bioinformatics analysis services to analyze off-target effects.?

These results show promise for age-related disease treatment in humans, which are often characterized by protein misfolds and accumulation. Activating the proteasome via gate opening could be a viable treatment for neurodegenerative diseases.
