The 10,000 Brains Project has commissioned the development of a roadmap to guide the rapid adoption of AI within?neurodegeneration research. The resulting insights, including recommended approaches for addressing the key gaps and opportunities, will be shared with researchers worldwide beginning this year. The roadmap will also inform the creation of the collaborative research networks needed to rapidly move the field forward." I can't wait to see what comes of this work and how it can accelerate research efforts to treat and potentially eliminate Alzheimer's, Parkinson's, and other neurogenerative disorders. If science is the pursuit of answers, then we must ask the right questions. For Alzheimer’s researchers, the question then becomes: How do we make the biological changes to the brain in order to reduce amyloid and tau? This translates into finding the right molecule for the disease and one that will breach the blood-brain barrier which protects the brain from toxic substances in the blood but can also keep potential treatments from reaching the brain. It means ensuring that patients participating in clinical trials have Alzheimer’s disease and not a different form of dementia. It also means understanding the stage of the disease in individual patients, as well as degree of pathology. The use of imaging tools has been instrumental in answering these questions, allowing scientists and researchers to look inside the brain of patients to not only confirm the presence of amyloid and tau but also provide a view of the efficacy of potential treatments. Projects like the Alzheimer’s Disease Neuroimaging Initiative, @ https://adni.loni.usc.edu and public-private partnership to develop a longitudinal study of normal cognitive aging, mild cognitive impairment (MCI) and early Alzheimer’s disease as a public domain research resource have provided tremendous insights into disease progression. It has required commitment by academia, industry and government, and a drive and determination by numerous scientists and researchers around the world. But there’s more that needs to be done. Innovation is about connecting dots. No one person or organization alone can advance science. It’s about seeing connections among things that are going on around us, experiments in academic labs as well as clinical trials. It’s also about seeing what’s possible and then striving toward it. The Women’s Alzheimer’s Movement at Cleveland Clinic recognizes that women are at the epicenter of the global Alzheimer’s epidemic and believe that cutting edge research on women’s brains is necessary to change the course of the disease. We raise money for critical women-based research and instigate the creation of women-based initiatives at leading scientific institutions, universities, foundations and organizations. We partner with leading researchers and non-profit organizations that share our vision and hope to create women’s Alzheimer’s research funds that finance high potential, gender-based scientific investigations. We are aggressively seeking answers to the question: Why are women disproportionately affected by Alzheimer’s and related dementias? Answering that question requires a sharp focus on what is particular to women’s biology, genetics, and lifestyle. While we recognize there is some gender specific research underway in labs around the country, we also know that there has not been a sufficient emphasis placed on determining the relationship between a women’s biological and genetic make-up and the fact that women are being diagnosed with Alzheimer’s, dementias and other neurological disorders at staggering rates. The myriad of gender specific factors like hormone levels, genetic composition, age-related variables, and lifestyle practices, provide a broad basis for the research that is essential to finding the answer that we hope will lead to breakthrough discoveries with far-reaching impact on the future of Alzheimer’s disease for women, men, and families everywhere. Since 2016, WAM at Cleveland Clinic has funded $4.65 million for 44 studies at 17 leading institutions and positioned its grantees to earn an additional $83 million more in government and foundation funding.

We have a great addition to your summer reading list!?Our 2023 Annual Report is out, full of science on the cutting edge of the fight against Alzheimer’s disease.?We are proud that it once again reflects a broad spectrum of diverse research.?Since our founding 20 years ago, we have sought to take on high-risk, high-reward projects reflecting a range of innovative but rigorously established hypotheses, and we supported over 100 such efforts last year alone.? The clinical benefits achieved by the new anti-amyloid drugs are encouraging as evidence that progress is being made, but we need to do better against amyloid as a target and against the myriad other targets this complex disease implicates. In 2023 alone, we provided grants to investigations focused on: ? Triggers and mechanisms of tau misfolding and propagation; ? Deeper understanding of the drivers leading APP cleavage towards beta amyloid production;? ? Mechanisms of synaptic and circuit dysfunction and support in AD; ? Neuroinflammation and the role of brain-resident and peripheral immune cells; ? Impact of immunization on AD pathology; ? Potential for gut microbiome interventions to affect AD brain pathology; ? Molecular pathways altered by lifestyle interventions like exercise, diet, sleep, and stress management; and ? Many other promising areas of research. ? We understand the importance of unconventional ideas in scientific breakthroughs; that's why we strive to be the funder that exceptional researchers turn to for support in exploring potentially paradigm-changing concepts.
