At ErebaGen we are always looking for ways to make current technology more accessible. A recent paper from Shufang Liang’s lab in Sichuan University describes a hybrid tool, CRISETR, that combines CRISPR/Cas9 and RecET-mediated Refactoring technologies to allow the rapid editing and refactoring of complex genetic networks in Streptomyces. Initially, there were a number of challenges associated with using CRISPR/Cas9 due to low efficiency, but advances such as these described by the Liang lab demonstrate how this tool can be hybridised with other technologies to broaden it’s range of applications. In our work we have found that it is challenging to compete with what Nature has already evolved, so tools such as CRISETR that allow the rapid editing of genomes and high-throughput screening can vastly accelerate our workflows. Here at ErebaGen we have developed our Forge platform to perform similar experiments, which is allowing us to use evolved pathways to create novel chemical compounds or complex intermediates in chemical processes. I thoroughly enjoyed reading this paper! If you're tackling a diverse set of synthetic biology challenges and want to generate large dataset, having reliable tools is crucial. While CRISPR is undeniably powerful, it still faces limitations, especially in high-throughput experiments. The solution from Shufang Liang group is a clever solution and you can see how this tool could be used in a number of problems. Stephen Quake, D.Phil. spoke at SynBioBeta on Tuesday, May 7, about the future of experimentation versus computation in science. Today, we do 90% experimentation and 10% computation to test our hypotheses. In the future, Stephen predicts that this will be inverted, with 10% experimentation and 90% computation. This statement really sparked my internal brainstorm. As we approach the inflection point to a predominantly digital twin world, how does the innovation ecosystem change? Does it become cheaper to innovate? Do we develop less technology to enable discovery, or more? Do we finally get over the hurdle of using genotype (+ myriad other factors) to accurately predict phenotype or real-world outcomes??I think it’s going to be an interesting road. Thanks to my fellow panelists Jason Kakoyiannis, Frank Jaksch, Chris Meaney and Robert Weinstein for our conversation on Plant Cell Culture at SynBioBeta 2024. It was great to have a lively conversation about how plants, on any scale (whole organism, callus or single cells), have the capability to be synthetic biology bioreactors right alongside their microbe friends.
