In a recent study (https://lnkd.in/gEPCgFHJ), Researchers from the University of Groningen established an in vitro co-culture model using ECIS to examine how real-time fermentation of non-digestible carbohydrates (NDCs) by gut bacteria affects gut barrier function. Lactobacillus plantarum WCFS1 and alginate-pectin were used to assess the impact of pectin fermentation on T84 gut epithelial cells. Encapsulation of L. plantarum in alginate gel proved effective, avoiding direct cell contact and creating anaerobic conditions. Testing with graded pectin loads showed increased protection of gut epithelium with higher pectin content, possibly due to enhanced short-chain fatty acid production. To assess the TEER (barrier function) of the gut cells, the researchers used Electric Cell-substrate Impedance Sensing (ECIS) where they were able to take measurements continuously in real-time while the cells remained incubated. Owing to their cargo including endemic functional micronutrients such as ascorbic acid, edible plant (or fruit) EVs are considered as an excellent platform for nutraceutical delivery. Proof generated through various studies suggest that their anti-oxidant and anti-inflammatory features pave the way towards broad horizons in clinics to tackle neural-, circulatory-, musculoskeletal-, and gastrointestinal-diseases. Yet, one big concern to exploit this potential of plant EVs is the lack of standardization in the ways they are isolated, stored, and preserved. In the search for enlightening the barriers to overcome, Theodora (Dora) Karamanidou and her colleagues preferred our TFF to isolate citrus EVs from their sources as an ideal method for standardization and large scale production. By using two different TFF cartridges within our portfolio, TFF-Easy and TFF-MVs, the group investigated the affects of the source and preservation conditions on the physicochemical characteristics as well as biological activity. In the end, the group succeeded in demonstrating that maturation of the use of plant EVs in high-end applications requires more in-depth investigations towards standardization. ?? I am happy to announce the publication of our latest article ?? "Evaluation of enzymatic protocols to optimize efficiency of bovine adipose tissue-derived mesenchymal stromal cell isolation" in npj Science of Food (top 10%). Sustainable food provision for a continuously growing human population is one of the major challenges for the next decades. Cultured meat represents one of the alternatives which is currently extensively explored. Yet, the most appropriate cell type, capable of long-term proliferation and myogenic differentiation, remains to be identified. This article focuses on considering bovine mesenchymal stromal cells (MSCs) a promising cell source. Within the context of cultured meat production, it is mandatory to maximize cell yield per tissue source. Although many enzymatic methods to isolate MSCs from adipose tissue (AT) have been described, cell yield has never been compared. In this study, we evaluate different isolation conditions. The highest cell yield in combination with a low population doubling time was obtained using Liberase at a concentration of 0.1% for 3h. MSC identity of the cells was confirmed by tri-lineage differentiation potential and cell surface marker expression. Subsequently, isolated cells were myogenically differentiated using 5-aza-2’-deoxycytidine and galectin-1. mRNA levels of the myogenic regulatory factors were increased.
