It doesn't matter how good your yields are. A prep is useless if it's full of garbage. But what's a main contributor to this problem? Scientists can't see these junk particles in upstream production. Not accurately and quickly, anyway. Instead, they have to rely on: - Quick methods that are inaccurate, or... - Accurate methods that are VERY slow This challenge destroys your ability to quickly test a variety of different conditions, then immediately focus on the parameters that reduce junk. However, I'm joining a company that fixes this challenge. I think it'll be transformational for AAV therapy. Stay tuned for the announcement on Monday! Recently there has been some chatter that there are no simple answers to characterizing the various species of AAV: specifically, full, empty, and partial. This is simply not true. AUC gives simple accurate precise quantification of 5 classes of AAV particles: aggregates, over filled, full, partial, and empty. This presentation given in 2020 summarizes the theoretical reasons why AUC can give simple answers while other methods cannot.

Latest App Note Released Now! ??? ? This application note explores the use of the SUPR-DSF to measure the thermal stability of three AAV serotypes: AAV2, AAV5, and AAV6. The SUPR-DSF measured the intrinsic fluorescence of each serotype during a thermal ramp to generate melt curves. In addition, a concentration dilution series was performed on AAV2 and AAV6 to determine the minimal sample requirements. All samples were performed at a well volume of 10μL in triplicate on a single 384-well microplate, emphasising the high-throughput possibilities of the instrument.
