"Today's dementia research investments are tomorrow's breakthroughs." 2023 was a landmark year for the Alzheimer's Association?, the world's largest nonprofit funder of Alzheimer's research. The organization invested $100 million in Alzheimer's research initiatives, making 2023 the largest single year investment since the Alzheimer's Association's founding. Over the last decade, the Alzheimer's Association has seen an increase in diversity of the projects being funded—research exploring alternative mechanisms of the disease and different approaches to treatment and disease management. The Women’s Alzheimer’s Movement at Cleveland Clinic recognizes that women are at the epicenter of the global Alzheimer’s epidemic and believe that cutting edge research on women’s brains is necessary to change the course of the disease. We raise money for critical women-based research and instigate the creation of women-based initiatives at leading scientific institutions, universities, foundations and organizations. We partner with leading researchers and non-profit organizations that share our vision and hope to create women’s Alzheimer’s research funds that finance high potential, gender-based scientific investigations. We are aggressively seeking answers to the question: Why are women disproportionately affected by Alzheimer’s and related dementias? Answering that question requires a sharp focus on what is particular to women’s biology, genetics, and lifestyle. While we recognize there is some gender specific research underway in labs around the country, we also know that there has not been a sufficient emphasis placed on determining the relationship between a women’s biological and genetic make-up and the fact that women are being diagnosed with Alzheimer’s, dementias and other neurological disorders at staggering rates. The myriad of gender specific factors like hormone levels, genetic composition, age-related variables, and lifestyle practices, provide a broad basis for the research that is essential to finding the answer that we hope will lead to breakthrough discoveries with far-reaching impact on the future of Alzheimer’s disease for women, men, and families everywhere. Since 2016, WAM at Cleveland Clinic has funded $4.65 million for 44 studies at 17 leading institutions and positioned its grantees to earn an additional $83 million more in government and foundation funding.

We have a great addition to your summer reading list!?Our 2023 Annual Report is out, full of science on the cutting edge of the fight against Alzheimer’s disease.?We are proud that it once again reflects a broad spectrum of diverse research.?Since our founding 20 years ago, we have sought to take on high-risk, high-reward projects reflecting a range of innovative but rigorously established hypotheses, and we supported over 100 such efforts last year alone.? The clinical benefits achieved by the new anti-amyloid drugs are encouraging as evidence that progress is being made, but we need to do better against amyloid as a target and against the myriad other targets this complex disease implicates. In 2023 alone, we provided grants to investigations focused on: ? Triggers and mechanisms of tau misfolding and propagation; ? Deeper understanding of the drivers leading APP cleavage towards beta amyloid production;? ? Mechanisms of synaptic and circuit dysfunction and support in AD; ? Neuroinflammation and the role of brain-resident and peripheral immune cells; ? Impact of immunization on AD pathology; ? Potential for gut microbiome interventions to affect AD brain pathology; ? Molecular pathways altered by lifestyle interventions like exercise, diet, sleep, and stress management; and ? Many other promising areas of research. ? We understand the importance of unconventional ideas in scientific breakthroughs; that's why we strive to be the funder that exceptional researchers turn to for support in exploring potentially paradigm-changing concepts. One of the areas in which the University of Exeter is working to make the world a healthier place is its research into Parkinson's: a brain disorder that causes slowness of movements, shaking, stiffness, and difficulty with balance and coordination. In 2022 the The Michael J. Fox Foundation for Parkinson's Research (MJFF) provided a ￡2.6 million programme grant to fund a research team, led by Exeter, for two studies to find new ways of understanding what happens in Parkinson’s before and after it is diagnosed, and to explore the role of genes in the evolution of Parkinson’s. This research is led by the Neurodegeneration Imaging Group (NIG) - a world-leading academic group that performs clinical research on the causes and mechanisms underlying neurodegenerative diseases - headed up by the Director, Professor Marios Politis. Today, on World Alzheimer’s Day, we reflect on the challenges faced by millions of patients and families affected by this devastating disease.? ? At?3D-PharmXchange, we are honored to support?Treeway?in its mission to develop groundbreaking therapies for neurodegenerative diseases. One of the key projects we're involved in is the development of?oral edaravone (TW001), which is currently being tested in?the ASURE study,?a?Phase 2a clinical trial?for?mild-to-moderate?Alzheimer's disease. This promising therapy has the potential to slow down disease progression by targeting oxidative stress, a critical component in the pathology of Alzheimer's. ? Recently, Treeway announced that they have successfully?completed enrolment for the ASURE study, marking a significant milestone in the development of TW001. This trial will evaluate the safety, tolerability, and potential efficacy of oral edaravone in patients with Alzheimer’s, and we are excited to be part of this pivotal journey.? ? Another great example of the value of mechanistic modeling. Applying treatment guidelines in the clinic can be a challenge when the patient population shares few commonalities with those studied in the clinic trials, as is the case with Alzheimer’s disease.?This paper demonstrates how the biophysical basis of a QSP model can help leverage clinical trial learnings to create treatment guidelines for the larger heterogeneous population. ??
