By repurposing a targeted chemotherapy drug called larotrectinib, an international research team successfully transformed treatment-evasive ‘cold’ tumours into 'hot' tumours that the immune system can attack ?? This breakthrough stems from a notable observation in clinical melanoma samples, which involves the nerve growth factor (NGF)—a protein that promotes the growth of nerve cells. Experiments using CRISPR-Cas9 gene-editing technology confirmed that NGF-deficient melanoma cells grew slower and allowed more immune cells, such as CD8+ T cells, to penetrate the tumours. Building on these findings, the researchers formulated a distinct genetic profile from NGF-deficient tumours that may be used to predict how well melanoma patients would respond to immunotherapy. Our Achievements: Development of new cancer treatment compounds We are excited to share a significant achievement of our N.N. Petrov National Medical Research Center of Oncology in collaboration with scientists from Peter the Great St. Petersburg Polytechnic University, ITMO University, and Perm State University. Together, we have developed and synthesized unique compounds that suppress the growth of malignant cells. One of these compounds, pyrrolothienopyrimidine, has garnered special attention due to its high biological activity. Initially, it was tested in vitro on cells and then in vivo on animals (mice). The studies showed that this substance effectively reduces tumor size without negatively affecting healthy organs. In the early stages of testing, we focused on colorectal cancer, breast cancer, and melanoma. The results were impressive: in the group of mice with the highest dose of the drug, the tumor size decreased fourfold. It is important to note that the substance also effectively impacts metastases, making it promising for the treatment of late-stage cancer. We have now moved to the next phase — testing the substance on human cancer cells. Preliminary results also confirm its effectiveness. This breakthrough opens new horizons in cancer treatment, providing more effective and safer alternatives to traditional chemotherapy.
