First paper is always special ?? I'm absolutely delighted to share that our comprehensive literature review on "Advancements in Nanoparticles for Drug Delivery, Therapeutic Applications and Computational Modeling" has been published! ???? This extensive review explores the development of nanocarriers for precise drug delivery and their potential to improve targeting and therapeutic outcomes. We analyzed how engineered nanomaterials can enhance drug potency while minimizing toxicity – a promising approach for cancer treatment. I'm grateful to have contributed my expertise in examining the psychological insights on patients' preference for nanomedicine over conventional chemotherapy. Incorporating this human-centric perspective adds valuable depth to our work. Kudos to my brilliant co-author Souhrid Sarkar for leading this review and bringing together our collective knowledge. I'm excited about the potential impact of our literature analysis in revolutionizing disease diagnosis and paving the way for targeted, potent, and focused cancer therapies. AlzeCure Pharma is presenting preclinical data with its lead drug candidate NeuroRestore ACD856 at the Alzheimer's congress AD/PD 2024, which will be held in Lisbon on March 5-9. The abstract and poster is titled ”ACD856 is a biased positive allosteric modulator of Trk-receptors - Enhances neurite outgrowthbut do not affect pain signaling” is by Pontus Forsell, Head of Discovery and Research at AlzeCure with co-authors Veronica Lidell, Azita Rasti, Gunnar Nordvall and Johan Sandin. The presentation includes study results showing that ACD856, the lead drug candidate in the NeuroRestore platform being developed with a focus on Alzheimer's disease, selectively appears to potentiate some signaling pathways, but not others. New preclinical data show that the substance stimulates the growth of nerve cell projections, which can promote nerve cell communication, but does not affect other functions, such as pain signaling. Previous studies have also been able to show that ACD856 improves memory, synaptic plasticity and has neuroprotective properties. “The selective effects of ACD856 are a strength for the continued clinical studies and show that substances from the project can have potent effects while maintaining a good safety profile,” said Pontus Forsell. “These positive data are very important for AlzeCure as we can now continue the development of this potential blockbuster in Alzheimer's disease where there is still a great medical need," said Martin J?nsson, CEO of AlzeCure Pharma. Dear Connections, I am absolutely thrilled to share a significant milestone in my research journey: our paper titled "The Power of Synergism: A Novel Drug Combination for Improved Therapy of Glioblastoma" has been accepted for publication in the esteemed journal Advanced Therapeutics! In this study, we explored the synergistic potential of combining disulfiram (DSF) and 7-ethyl-10-hydroxycamptothecin (SN-38) in targeting glioblastoma (GBM) through the PI3K/AKT/mTOR pathway. What's truly remarkable is that this combination demonstrated a highly potent effect against GBM, even at nano-molar concentrations, surpassing the efficacy of the gold-standard treatment, TMZ, which typically acts at a micro-molar concentration. We were excited to collaborate with Tessara Therapeutics to co-present a poster at ELRIG Drug Discovery, London, October 2024. The poster, “Development of iPSC-derived 3D human brain micro-tissues for drug discovery applications” generated iPSC-derived RealBrain? micro-tissues to model Alzheimer’s disease. Key takeaways: ? 3D neural micro tissues were generated on Tessara’s automated platform using iPSC derived neuronal stem cells from Axol Bioscience and proprietary biomaterials. ? Tessara’s ADBrain micro-tissues are generated following exposure of cells (primary human cells in this panel) to a proprietary Aβ formulation that results in neurodegeneration consistent with that seen in Alzheimer’s disease. ? This models key pathological features of the AD brain including; Aβ42 deposition, loss of neural networks, hyperphosphorylated Tau, dystrophic axons and senile plaques. ? This ADBrain model is more susceptible to cell death as compared to the healthy brain. The conclusion states we successfully demonstrated proof-of-concept for the development of iPSC- and patient-derived 3D human brain micro-tissues. This work will pave the way for the development of protocols for “clinical trial in a dish” studies to help accelerate personalized medicine approaches for neurological conditions and will help drive innovation and knowledge translation. It will also significantly de-risk drug discovery pipelines by reducing cost and providing earlier insights into drug neurotoxicity and therapeutic efficacy. With current changes in the regulatory landscape, this work will also foster the adoption of non-animal-based screening approaches across the sector. ?? Exciting Collaboration Alert! PROTEINA Teams Up with Emory University School of Medicine for Clinical Validation of PPI Technology in AML! ?? We're thrilled to announce that PROTEINA, a leader in protein-protein interaction (PPI) technology, has embarked on a pivotal clinical validation project in collaboration with Emory University School of Medicine, Atlanta, USA. Our partnership focuses on enhancing the precision of drug response predictions in acute myeloid leukemia (AML) patients using our groundbreaking PPI PathFinder BCL2 Dx offering. This tool aims to analyze protein interactions with high sensitivity and predict responses to the targeted anticancer drug, venetoclax, more accurately. Following successful clinical validations in South Korea in collaboration with Seoul National University, which will soon be published in the prestigious Nature Biomedical Engineering journal, we are expanding our trials to the US. Starting next month, our equipment and reagents will be shipped to labs at Emory to analyze over 400 clinical samples for validating and expanding the applications of this technology.
