How do you turn a Llama into a Human? When you look at the sequence of camelid (e.g. Llama) VHH, you could be forgiven for thinking they are the same as a human VH – in fact many people do. However, the human immune surveillance system is not so easily fooled, so before making your VHH wonder lead into a wonder drug it is necessary to modify its sequence to resemble a human protein as much as possible – without changing it’s “drug-like” properties. Not so simple, as VHH are more complex than first appearances and even minor changes in disparate regions of their sequences can have profound effects. Happily, there are a number of ways to address this and although each method has its caveats, with the right expertise and technologies it’s often possible to humanise a VHH and maintain its mechanism of action and developability.
