In the small Syracuse lab of Zetagen Therapeutics, cancer researchers believe they’ve uncovered a new use for a common anti-overdose drug that eases the suffering of people with breast cancer that has metastasized into their bones. Scientist Bryan Margulies, who helped start the Upstate Cancer Center, has been experimenting for years with a modified form of naloxone. The parent drug, commonly known as Narcan, has been approved since 1971 to reverse the effects of opioids. It turns out that modified naloxone has promise as a miracle worker, of sorts, for people with breast cancer that has spread into bones, Margulies said. PAC-1, a drug originally developed by scientists at the University of Illinois Urbana-Champaign, induces programmed cell death in cancer cells. Notably, it halted tumor growth in five participants with?neuroendocrine cancers, with two of these patients experiencing tumor size reduction. This compound also demonstrated: ?? Selective cytotoxicity for cancer cells, sparing healthy ones. ?? Innovative mechanisms: Inhibiting thioredoxin reductase, inducing ROS accumulation, and promoting apoptosis. ?? Antiangiogenic effects: Stopping tumor blood vessel formation in zebrafish models. ?? Superior efficacy: Achieving 82% tumor inhibition in mouse models, far outperforming cisplatin’s 29%.
