Recently, I read two papers on lymphatic filariasis (LF) discussing Cofactor-independent phosphoglycerate mutase (iPGM) as a potential drug target for LF, published in Oxford Academy-2024 (PMID:37481257) and Bentham Publications-2023 (PMID:31244425). I'm glad to have discovered this back in 2015. Here are some insights from my original article if you're interested: https://lnkd.in/eTdnyXs3. We delved into the structural stability of Wb-iPGM through molecular dynamics simulations, shedding light on its interaction with cofactors 3-phosphoglycerate (3PGA) and Mn2+ ions. Imide-like compounds provide the scientific background for discovering and developing PROTACs and molecular glue drug compounds, two innovative strategies in targeted protein degradation. Amazing how the puzzle of 30 (thirty) new #HCAR2 structures when pieced together by Asli (Aslihan Shenol Iliyaz), Ricardo Tenente, Michael Lückmann, and Thomas Frimurer shows the whole beautiful picture of the electrostatically guided, complex agonist binding path into a totally occluded orthosteric site in the middle of the receptor; how an #agoPAM surprisingly works by controlling the dynamics of the entry/exit gate; and how a two-way allosteric mechanism connects G protein with agonist binding. G protein binding ALONE stabilizes ALL major conformational changes - including extracellular loop 2b - except only the induced-fit anchoring of agonist in the orthosteric pocket.
