Anti-PD-1 and anti-PD-L1 antibodies are key immunotherapies for cancers- choosing the right antibody is crucial, especially for advanced ones. This protocol describes an imaging-based assay developed to compare the performance of various anti-PD-1 and anti-PD-L1 antibodies. Existing assays overlook PD-1’s role in the immune synapse, a critical factor in its biology. The new assay quantifies the ability of antibodies to remove PD-1 from the immune synapse and correlates this with increased cytokine secretion, demonstrating its potential for personalized medicine.
