Proud to share our three-year-long effort to systematically understand the response mechanisms of CDK4/6 inhibitors (CDK4/6i). https://lnkd.in/gRUcfuwW - We trained an interpretable AI model of palbociclib (a CDK4/6i) response on cell-line data. We applied our model to patient and PDX data to validate its accuracy. We found that the model specifically learns CDK4/6i response. - We interpreted the model to identify 8 core protein assemblies that integrate rare and common alterations across 90 genes. Validated assemblies relate to cell-cycle control, growth factor signaling, and histone regulation. - We experimentally validated a histone transcriptional assembly to show overexpression of KAT6A and TBL1XR1 causes resistance to palbociclib. - This work was co-led by Dr. Sungjoon Park, Dr. Erica Silva, and Dr. Trey Ideker.
