One approach is the use of long-acting injectables (LAIs), which can release an active pharmaceutical ingredient over weeks to months. These formulations often utilize aqueous suspensions with crystalline drug particles engineered to maintain a predefined plasma concentration profile by controlling the particle sizes. The production of nanosuspensions typically involves wet bead media milling reducing larger drug particles to smaller ones through mechanical friction in an aqueous stabilizer vehicle. However, the inherent thermodynamic instability of these systems can affect their long-term stability. To counteract issues like agglomeration and Ostwald ripening, stabilizers such as surfactants and polymers are added preventing inter-particle forces through electrostatic repulsion and steric stabilization. The choice and concentration of stabilizers depend on the specific formulation and the physicochemical properties of the drug, making the selection process complex and often reliant on trial and error relying on empirical data. In this talk, recent insight into the stability behavior of suspensions, with respect to the particle size growth, and also resuspendability are discussed, as well as some of the initial learning applying machine learning (ML) to suspension formulations to understand these systems even further.
