Investigating how conjugation chemistry influences targeted nanoparticles: in their latest work, Michael Zaleski, Jake Brenner at the University of Pennsylvania and collaborators demonstrated that the commonly used conjugation chemistries for antibody-targeted nanoparticles play a direct role in activating the complement cascade of plasma proteins. Z-alkenes are common structural components in both chemistry and biology. As such, the photoisomerization of E-to-Z alkenes has various applications in organic chemistry, polymer chemistry, and medicinal chemistry. In a new The Journal of Organic Chemistry study, researchers from Tokyo University of Science developed a new method for E-to-Z alkene photoisomerization using a recycling photoreactor. This method features an immobilized photosensitizer and high-performance liquid chromatography which recycles the samples, thus improving efficiency. This method can lead to more green and sustainable production of various organic compounds, including pharmaceuticals. Congratulations to Dr.Kridsadakorn Nook Prakinee,research team and collaborators of Prof. Dr.Pimchai Chaiyen group for publishing in Angewandte Chemie, a Nature Index journal (IF 16.6). Dealing with native flavin-dependent halogenases presented challenges, so they sought more robust enzymes. The team addressed the question with mechanistic approach and biocatalysis goal. The study revealed that certain flavin monooxygenases could be redirected to enzymatically generate hypohalous acid, a key halogenation intermediate. Through stopped-flow experiments and computational analysis, they uncovered their mechanisms. Eventually, they developed a new chemoenzymatic iodination method that is efficient and sustainable. My last postdoc project has just been published - a collaboration with Applied Materials (fei wang, Pravin Narwankar, Kai Zheng, Miaojun Wang) and Purdue University (Emily Benson, Shradha Bhalla, Lynne Taylor) studying the impact of atomic layer coating on dissolution performance of copovidone-based amorphous solid dispersions. In previous studies, the ALC coating did not impede dissolution performance of HPMCAS-based ASDs where release was rapid, and could slow matrix crystallization where release was slower. In copovidone-based ASDs, due to the conflict of the gelation release mechanism with the rigid coating, release rates reduced when coated, highlighting the importance of rapid hydration kinetics on ASD drug release. By formulating the coated ASD powders with superdisintegrants in the tablet formulation, rapid release was again achieved, mitigating the release rate reduction. Happy to share our publication recently appeared in Chem Catalysis on a new biocatalytic strategy for the regio- and stereoselective C(sp3)��H functionalization of indolines via carbene transfer chemistry with engineered heme enzymes! In this work, we demonstrate the development of CYP119-based carbene transferases for the selective alkylation of three different C(sp3)��H sites in indoline substrates with high enantioselectivity, high catalytic efficiency, and regiodivergent selectivity. These systems thus offer a unique and convenient route for diversification of these important organic scaffolds via direct C(sp3)��H functionalization. Using enzyme variants with complementary regioselectivity, a tandem inter- and intramolecular C(sp3)-H carbene insertion sequence could be orchestrated to afford a polycyclic indoline-containing motif found in drugs. Through mechanistic and computational studies, we shed light into the mechanism of this abiological reaction and into structural features responsible for the enzyme-controlled regiodivergent selectivity. This is the first-ever combination of polymerization-induced self-assembly (PISA) with cationic polymerization of 2-oxazolines��offering a straightforward, one-step method to create stable nanoparticles directly from monomers. ?????? Mind the Surface Coverage - What to Consider When Designing Your Catalyst Characterization Experiment? What I often see in catalysis research papers is a statement like: "No CO is found on the surface in the temperature-programmed experiment above 150属C. Therefore, surface CO is not a relevant surface intermediate during the reaction at 200属C because it is not stable above 150属C." This is very often a WRONG statement. Thermodynamics and kinetics determine the degree of coverage on the catalyst surface. Let's stick with the CO example and look at how CO covers a platinum surface. To get a rough idea, we really do not need to do any experiments but just some calculations. What we need to know is: -The CO gas pressure -The Temperature -A rough estimation of the energy of adsorption on the surface in question With some simplifications, this is all we need to get a decent idea of how the coverage of CO is on platinum. In the top figure, you see the coverage of CO as a function of temperature for a variety of pressures. The figure below shows you what would change if the bindinge energy of CO would be different. Now we can go back to why the statement is often wrong. Generally, we perform temperature-programmed experiments like CO-TPD without CO in the gas phase. So the CO pressure is at best very, very small. CO as an adsorbate is not stable under these conditions. And if your CO-TPD experiment gives a desorption peak at higher temperatures, this is the kinetic barrier. However, under your reaction conditions, when CO is present in the gas atmosphere, you may easily have CO covering the surface. We have to be very cautious about this fact, especially if we look at in-situ experiments that operate at lower pressures than the catalytic reaction that we target. For example, working with "Ambient Pressure" XPS, which usually operates around 1 - 10 mbar total pressure, will show fewer and different surface species. Maybe important surface reactions even cannot be induced under such conditions. This seems like physical chemistry 101, but in the heat of the experiment, such things are often overlooked. The latest publications regarding the preparation and properties of polypeptides and their blends with different polymers have been assessed.
