Our latest publication with Pfizer, “Building Efficient Diastereo- and Enantioselective Synthetic Routes to trans-Cyclopropyl Esters for Rapid Lead Scale-Up," has been selected as an Editor’s Choice in American Chemical Society's Organic Process Research & Development! Led by Pfizer scientists, this work introduces the first example of a sulfoximine-driven Johnson–Corey–Chaykovsky reaction for the enantio- and diastereoselective synthesis of trans-cyclopropanes, expanding the synthetic toolkit for these valuable drug-like structures. Additionally, we developed a novel scalable process route that optimizes a ring-opening/intramolecular phosphate transfer and displacement cascade, enabling highly efficient stereocontrolled synthesis from chiral epoxides. By addressing key challenges in cyclopropane synthesis, our collaborative research accelerates lead optimization and provides medicinal chemists with practical, scalable solutions for drug discovery. As a data-driven small molecule discovery partner, we're excited to share that our recent research has been published in American Chemical Society's Journal of Chemical Information and Modeling! In this study, we introduced a novel "multiclass fold-ensembled classification model" to predict the minimum number of synthesis steps required for small molecules. This method addresses challenges like data imbalance and provides a more accurate and flexible prediction of synthetic feasibility, with validation showing our model outperforms six benchmark models. By moving beyond predefined thresholds, we aim to contribute to more informed decisions in early-stage drug development.
