I am thrilled to announce our latest publication, "Real-Time Monitoring of Higher-Order Structure of RNAs by Temperature-Course Size Exclusion Chromatography and Microfluidic Modulation Spectroscopy," now available in Analytical Chemistry. This work is the result of a fantastic collaboration between the talented teams between #Genentech Shijia Tang, Lance Cadang, Bifan Chen, Jenny Wang and Kelly Zhang and RedShiftBio team Eugene Ma, Valerie Ivancic Collins, Ph.D., Scott Gorman, Richard Hailin Huang, Ph.D. and Maria MA, Ph.D.. You can access the full publication here: https://lnkd.in/gvfCnhRP In this study, we took an in-depth exploration of the biophysical characterization of the higher-order structure (HOS) of RNAs. By developing two complementary native analytical techniques—Size Exclusion Chromatography (SEC) and Microfluidic Modulation Spectroscopy (MMS)—we successfully monitored changes in RNA HOS and thermal induced dissociation of RNA aggregates in real time. We established a beautiful correlation between the separation and spectroscopic results. We believe that the combination of SEC and MMS provides a comprehensive toolkit for the biophysical characterization of RNA HOS under native conditions, offering valuable insights for optimizing candidates and screening formulations in RNA therapeutic development. Additionally, Richard Hailin Huang, Ph.D., Valerie Ivancic Collins, Ph.D., and I have contributed to a #MyACSCover story for our study. We depicted a thermal ramping MMS setup on the ground, symbolically dissociating double-stranded RNA in the flow into two #AuroraTX in the night sky. Enjoy the story!

?? Buffers Matter More Than You Think Pick the wrong buffer, and your protein might not behave the way you expect. But how do you measure those structural changes with real confidence? We used MMS (Microfluidic Modulation Spectroscopy) to analyze lysozyme in water, PB, PBS, and Tris. The results were clear. Tris had the biggest impact, followed by PB, then PBS. MMS detected these shifts with far greater sensitivity than traditional methods. No more guesswork, just data-driven buffer selection. Check out the full app note below. ??
