We are proud to announce that Rinnie Rodenius has joined Polymorph as a Co-Owner and Head of Operations! Rinnie is a highly experienced plant biologist with an extensive background in advanced tissue culture techniques and laboratory management. While studying TC under Dr. Michael Kane at the University of Florida she assisted in successfully eradicating viruses from commercially relevant species through meristem dissections and was awarded for her extensive contributions to endangered species conservation, such as the incredibly rare Ghost Orchid (Dendrophylax lindenii), by the Society For In Vitro Biology (SIVB). After graduating with honors she went on to complete her Masters of Science in Plant Biology and Conservation at Northwestern University and the Chicago Botanic Garden. Following her studies, Rinnie led Ball Horticulture's HQ tissue culture lab, servicing ornamental customers across 5 continents. She refined her skillsets here while building Ball's Clean Stock Department, spearheading contamination reduction R&D initiatives, and also served on the committee for the design and build of their new Tissue Culture Headquarters in Chicago. Rinnie has since continued her work in the cut flower industry, holding production and R&D department management roles for Conception Nurseries, one of the largest tissue culture providers in the United States. Here she pioneered their clean stock, initiation, and genetic storage protocols to support their continued expansion and success. Rinnie is an extremely experienced and extraordinary tissue culturist focused on producing and preserving the highest quality plants possible to support plant breeders, nurseries, and growers. Check out these results from our team’s peer reviewed tissue culture research! The word auxin is derived from Greek and means “to grow or enlarge.” Auxins are a group of plant growth regulators (hormones) that play a pivotal role in cell expansion and differentiation, root and stem tropisms, apical dominance, as well as flower and fruit development. In the realm of micropropagation, auxin is most often utilized to reduce time to root formation and increase root production in propagules. When establishing a media recipe the type and concentration of auxin, as well as the interaction with other growth regulators in the medium, need to be optimized depending on the species and objective. Although an important media component, it can cause issues such as explant stress, excessive callus formation, and worst of all, mutagenic effects (if the goal is to maintain true-to-type plants). Differences in root production and callus formation can be clearly observed based on the treatments in the picture provided. What are apical meristems and why do we care about them in plant tissue culture? Meristems are located at the tip of every shoot (and root) where cells are actively dividing to generate new tissues. These bundles of cells are only a few millimeters wide and not yet connected to the plant’s vascular system so pathogens are less likely to be translocated here. The concept of meristem culture was first developed in the early 1950s by George Morel and Claude Martin to remediate Dahlias from production inhibiting pathogens. Dissecting apical meristems is recommended to achieve the cleanest explant possible during the initiation stage. It is important to note that a properly dissected meristem under a microscope will be smaller than a grain of sand and barely visible to the naked eye following initiation.
