Sometimes it's not the spectrophotometer but the solvent. 🤯 Water, methanol, chloroform— name a solvent and learn its UV cutoff before using it for a measurement which demands that everything be optimal. Take CD, circular dichroism. Countless experimenters have been frustrated by poor data below their preferred lower wavelength limit. It's easy to blame the instrument. But look first at your solvent. No instrument can overcome too little light making it out of a highly absorbing solvent. 🔬Important Examples: • Water <185 nm • Methanol 195 nm • Tris 100mM 195 nm • Urea 4M 210 nm You want your protein to be happy, of course. But you also need the solvent to not absorb all the photons!
