Our #metaboliteofthemonth N-acetylneuraminic acid (Neu5Ac) is part of the kidney’s glomerular filtration barrier. This sialic acid, produced in the hexosamine biosynthetic pathway, attaches to glycans on the surface of glomerular cells. Glycoproteins and glycolipids sialylated with Neu5Ac form part of a the glycolyx, a negatively charged, gel-like layer that acts like a sieve, preventing proteins from leaking out of the blood into urine. At our latest journal club, Dr. Masoumeh Dorrani reviewed a study published in the Journal of Chromatography B. The research developed and successfully validated a sensitive, specific, and fast U-HPLC–MS/MS assay for quantifying MPA and its major metabolites in plasma and urine. This assay includes automated sample preparation using a robotic system with 96-well plates. The study highlights the advantages of LC/MS assays in measuring immunosuppressants due to their high specificity. Key achievements include the complete separation of MPA, MPAG, and AcMPAG, as well as relatively short analytical run times of just 2.3 minutes between injections. Congratulations to CEO Liz O'Day who has been nominated as a candidate for Medical Sciences Trustee for United States Pharmacopeia (USP). USP was founded in 1820 by physicians who wanted to protect patients from poor-quality medical prepartions. It has grown into a nonprofit body of 300 delegates representing state and national associations and colleges of medicine, nursing, pharmacy, industry and agencies of the federal government. #MetaboliteoftheMonth alert! Say hello to N-acetylneuraminic acid (Neu5Ac) -- the only sialic acid synthesized de novo in humans. Sialic acid-containing structures on cell surfaces have important physiological functions, like keeping blood flowing smoothly. Sialic acids are negatively charged nine-carbon monosaccharides. Neu5Ac is synthesized in the hexosamine pathway and converted to CMP-Neu5Ac in the nucleus. Then in the Golgi, sialic acid modifies the ends of glycoproteins and glycolipids destined for the cell surface. Charge repulsion between sialylated red blood cell surfaces and sialyated blood vessel surfaces (endothelial glycocalyx) prevents red blood cells from sticking.
