#onlywithHCR: Visualizing Lymphatic Endothelial Cell Formation in Mammalian Vasculature??? A groundbreaking study from Lupu et al. challenges traditional views on the formation of lymphatic endothelial cells in mammals. Previously, it was believed that these cells emerged from a common pool of progenitor cells through a process of differentiation. However, their research reveals that lymphatic endothelial cells are actually derived from a specialized population of progenitors. By using #HCRtechnology, the authors were able to validate their scRNA-seq analysis and visualize endothelial progenitors co-expressing master regulators, Etv2 (endothelial) and Prox1 (lymphatic) in whole-mount mouse embryos. #onlywithHCR: HCR? Technology Enhances Butterfly Brain Gene Mapping ?? In this study on?Bicyclus anynana, HCR? technology played a pivotal role in visualizing gene expression in the larval brain. By optimizing the protocol, Banerjee et al. were able to map the expression of ten specific genes in whole brain tissue with single-cell resolution. The precision of HCR? imaging allowed them to capture detailed expression domains of genes like optomotor blind (omb), Synapsin, and SNAI2, which are crucial for understanding brain functions and behaviors. A study on Drosophila by Bayala et al. highlights a novel protocol using HCR? technology to study RNA expression in the fragile and impermeable tissues of the Drosophila epidermis during pupal development. Erick Bayala and his co-authors describe a method for dissecting tissue and detecting RNA across various stages, including early and late pupal stages, as well as larval imaginal discs. Powered by HCR? imaging, this approach allows for the study of dynamic gene expression patterns throughout the pupal development process, overcoming the challenges of tissue fragility and impermeability.
