Assay interference caused by heterophilic antibodies (HA) such as human anti-mouse antibodies (HAMA) can cause false-positive and false-negative results. IVD manufacturers often use animal serums or purified animal IgG to reduce this interference, however, these must be used in high concentration and can only block cross-reactive antibodies from a single animal of origin. TRU Block™ (Cat# A66800H) is a unique heterophilic antibody blocker that can be used at concentrations 10-100x less than animal IgG and it can remove many different types of heterophilic interference including HAMA and rheumatoid factor (RF) using a completely new approach. A new report released last year from the American Cancer Society (https://hubs.la/Q038Jlr30) reveals that cervical cancer is increasing in women in their 30s and 40s, most likely due to a lack of screening. More than half of women diagnosed with cervical cancer have either never been screened or have not been screened within the past five years. Traditional pap smears, which miss more than 40% of abnormalities, are being replaced by new HPV viral DNA tests. These tests are based on PCR techniques and detect up to 14 different HPV types within a single assay in approximately 60 minutes. Some tests allow for patient sample self-collection, eliminating the need to visit a doctor and receive a pelvic examination. Meridian’s Lyo-Ready Genotyping Direct qPCR Urine Mix is designed for fast, precise, and reproducible allelic discrimination and cluster separation, ideal for developing self-collection, PCR-based HPV-type tests. Its advanced formulation enables highly sensitive amplification of SNPs and other point mutations from urine samples and overcomes inhibitors such as urea and nucleases that can damage DNA and inhibit PCR reactions. It can be used in a wet format or lyophilized to create ambient-temperature stable assays, making it ideal for multiplexing in fast cycling analyzers and point-of-care (POC) devices. Cancer is the second leading cause of death worldwide, however, only 25% of cancers are detected via screening. The remaining 75% are detected when the patient is symptomatic. As mentioned in an article in the Journal of Translational Medicine (https://hubs.la/Q038w_FD0), liquid biopsy holds the promise for quicker and more accurate early-stage cancer detection across a wide range of cancer types using a single blood test, or other specimen type. The most common technology used in liquid biopsy is qPCR, followed by droplet digital PCR (ddPCR) and next-generation sequencing (NGS). There are several distinct advantages to qPCR, including cost-effectiveness, rapidity, and accessibility. To simplify the development of liquid biopsy assays, Meridian offers master mixes formulated to overcome inhibitors found in blood, urine, saliva, or stool samples, delivering ultra-sensitive detection down to single copies of DNA. They can be used on crude samples or extracted DNA/RNA, offering a flexible workflow, and they are designed to be used as a wet mix or dried down by air-drying or lyophilization to create ambient-temperature stable assays. Did you know qPCR is playing a critical role in advancing liquid biopsy? As the demand for non-invasive, high-precision diagnostics grows, qPCR stands out for its speed, sensitivity, and scalability, making it ideal for known targets in companion diagnostics (CDx) and minimal residual disease (MRD) monitoring. With real-time detection, qPCR delivers fast, actionable results in hours, accurately identifying low-abundance ctDNA, miRNA, and genetic mutations. Its high throughput and efficiency make it ideal for routine screening, seamlessly integrating into assay workflows. Meridian’s Specimen-specific™ inhibitor tolerant qPCR master mixes provide ultra-sensitive detection, accurately identifying single-digit copy numbers of targets like ctDNA from blood, saliva, urine, or stool samples. Designed for high performance with complex biological samples, they enable reliable, reproducible results, making them an ideal choice for liquid biopsy assays. Meridian’s Low DNA Taq HS is a highly purified, chemically modified hot start Taq that is ideal for workflows requiring room-temperature reaction setup. In high throughput settings where large-volume sample preparation may take an extended period of time, hot-start polymerases are essential to prevent nonspecific amplification and the formation of primer dimers. Free from animal-derived compounds, Low DNA Taq HS minimizes the potential for background contamination or DNA contamination, which is especially important for the detection of bacterial targets such as E. coli. The enzyme is completely inactive at room temperature and requires a 10-minute incubation at 95°C to release its functional activity.
