I am so delighted to formally announce Longevity Global's east coast summit in June, the Aging Code Summit. It's a 2-day event with a uniquely structured summit as the first day and a pitch competition as the second. It's created in partnership with Noriko Yokoi, Ph.D. and Daniel Dacey (Mindvyne) Day 1 - The first day, our AI in Longevity Summit won't just be a bunch of lectures, we've curated panels of the top visionaries in Longevity biotech and you get to watch what happens as they discuss different aspects of the rapid progress of longevity innovation, centered around the impact of AI. I've posted some of the speakers below and we're likely to have some surprise guest speakers as well. Day 2 - The second day will be a pitch competition featuring innovative startups. You will get to meet top investors and entrepreneurs in the field and hear from our finalists before a winner is selected. New study digging into who has side effects with Lilly's anti-amyloid antibody therapy for Alzheimer's and why. This could allow us to predict and avoid side effects. The worsening of side effects in people who have the genetic risk factor, APOE4, is hypothesized to be due to their greater amyloid burden. Starting therapy earlier, before they have so much amyloid accumulation, could be a good strategy for lessening side effects. There are serious side effects (brain bleeding and swelling (ARIA)) with Lilly's newly approved anti-amyloid therapy, Donanemab, in about 20% of people. There were 6 factors that predicted who had side effects. The largest risk factors were: -APOE4 carriers -high blood pressure -siderosis (the accumulation of iron deposits in the brain's surface layers) Read the study: https://lnkd.in/g8cPUbZN
Donanemab, an amyloid-targeting therapeutic,?was recently approved in July 2024. Third such therapy to be approved brought hope with evidence that stopping treatment when amyloid plaques were removed is possible. This critical article summarizing the safety issues (ARIA - amyloid associated imaging abnormalities) associated with this treatment highlights that much much more needs to be developed in this space. Namely, the analysis from TRAILBLAZER-ALZ and ALZ 2 randomized clinical trials relveal: ?? Higher ARIA Risk: Donanemab increased ARIA-E (20–24%) and ARIA-H (27–31%) cases. ?? APOE ε4 Influence: APOE ε4 carriers, especially homozygotes, faced higher ARIA risk. ?? Early Onset: Most serious ARIA events occurred within the first three infusions. ?? Modifiable Risks: Elevated blood pressure increased ARIA risk; antihypertensives reduced it. ?? Monitoring Matters: Rigorous safety monitoring is crucial during treatment.
