    Can ketone bodies help clear misfolded proteins linked to neurodegenerative diseases? A new study suggests that β-hydroxybutyrate (βHB)—a key ketone body—plays a direct role in protein clearance and brain health. Key findings: → βHB selectively targets misfolded proteins, including amyloid-β, a hallmark of Alzheimer’s disease. → This protein-clearing effect was confirmed in vitro, ex vivo, and in vivo using mouse and nematode models. → βHB's effect on protein solubility is not dependent on covalent modification, pH, or solute load. → Ketone esters delivered to mice led to reduced brain protein aggregation. These findings suggest that ketosis may promote proteostasis (protein homeostasis), acting as a natural “janitor” for damaged proteins. This mechanism could be leveraged for therapeutic development in aging and neurodegenerative diseases (NDDs). ?? A new systematic review and meta-analysis of 20 RCTs suggests minimal impact on key CVD risk factors, challenging long-standing concerns about red meat and heart health. Key findings: → No effect on total cholesterol, HDL, triglycerides, or blood pressure. → Slight LDL increase (+2.7 mg/dL), but this disappeared when one study was removed. → No significant impact on apolipoproteins or VLDL-cholesterol.     Autism Spectrum Disorder (ASD) affects over 1 in 100 children globally and is associated with immune system changes, inflammation, and gastrointestinal issues. Abnormal gut flora is linked to ASD symptom severity. The ketogenic diet has shown promise in ASD improving symptoms by raising blood ketone levels, which may have neuroprotective effects. An interventional pilot study examined the effects of a modified ketogenic diet in seven children with ASD over four months. The diet was limited to 20-25 grams of net carbohydrate per day and included MCT oil supplementation. The study aimed to assess changes in microbiota, inflammation markers, and microRNAs. Key findings after 4 months on the ketogenic diet: - Decreased plasma levels of proinflammatory cytokines IL-12p70 and IL-1b - Increase in gut microbiota diversity - Increased expression of butyrate kinase in the gut - Reduced brain-derived neurotrophic factor (BDNF) levels (The authors suggest this might be due to more BDNF being used in the brain, reducing its amount in the blood) - Altered levels of brain-derived -associated microRNAs in plasma These results suggest that the KD may improve outcomes for those with autism spectrum disorder by reducing inflammation, correcting gut microbial imbalances, and affecting the BDNF pathway involved in brain activity.     Post-Traumatic Stress Disorder (PTSD) is a complex mental health condition often accompanied by metabolic disfunction. Current treatments, such as psychotherapy and medications, have limited effectiveness for some patients. This feasibility study explored the potential of a 4-week ketogenic diet supplemented with exogenous ketones (KD-KS) as a dietary intervention for adults with PTSD. The study focused on ketosis maintenance, side effects, and acceptance. Three of four eligible patients were recruited, with two completing the full intervention. Key Findings: ??Achieving and Maintaining Ketosis: All participants reached ketosis (BHB ≥ 0.5 mmol/L) within the first two days, and ketosis was maintained for 87% of the intervention period. ??Adverse Reactions: No serious adverse reactions were reported, though mild to moderate side effects (e.g., headache, fatigue) were reported on 70% of intervention days. ??PTSD Symptoms: The PTSD Checklist (PCL-5) scores decreased by 20 points (from 70 to 50) and 10 points (from 50 to 40) in two patients over the 4-week period, indicating clinically significant improvements. ??Quality of Life (QoL): Improvement was observed across multiple QoL subscales, with six of eight subscales improving for one patient and three for another. This pilot study is the first to investigate a ketogenic intervention for people with PTSD, showing rapid attainment of ketosis, patient compliance, and potential improvements in PTSD symptoms and QoL.     Can the ketogenic diet improve our dreams? This research focused on evaluating the effects of a very low-calorie ketogenic diet on sleep quality among women with overweight or obesity. Engaging 324 participants in a very low-calorie ketogenic diet for 31 days, the study assessed changes in both body metrics and sleep quality, utilizing the Pittsburgh Sleep Quality Index questionnaire for sleep evaluation. Results indicated that alongside notable improvements in anthropometric measures and body composition, the diet significantly boosted sleep quality. Correlations were observed between improved sleep quality and variables such as BMI, waist circumference, fat mass, and levels of high-sensitivity C-reactive protein (hs-CRP), with a reduction in fat mass being a primary indicator of enhanced sleep quality post-diet. The findings suggest that a very low-calorie ketogenic diet can beneficially affect obesity management and sleep quality in women with overweight or obesity, underscoring the role of sleep quality improvement in obesity treatment plans.
