A lively and informative presentation by IonOpticks’ own Dr Daniela-lee Smith, PhD at #USHUPO2025 on how to improve proteome coverage and reproducibility in large-scale analyses using packed emitter columns. An innovative method for resolving the molecular complexity of cellular structures has emerged: Filter-Aided eXpansion Proteomics (FAXP). Developed by Dong et al. at Westlake University, FAXP integrates hydrogel-based tissue expansion techniques with mass spectrometry to advance spatial proteomics research. Validation of the workflow was conducted across digested sections of nuclei, cells, and blank controls, analysed using an Orbitrap Astral interfaced with a Vanquish Neo, utilising an Aurora Elite XT for peptide separation. The results demonstrated a 14.5-fold increase in volumetric resolution, an 8-fold rise in peptide yield, and over 255% more protein identifications compared to previous methods. FAXP allows researchers to pinpoint regions of interest with precision while cutting sample preparation times in half. This makes high-resolution proteomics more accessible and paves the way for applications in cancer research, biomarker discovery, and precision medicine. Learn more: https://bit.ly/3ES1HwJ
