Congrats to Ben Niu and his crew at BMS on this end-to-end pipeline for bi- and multispecific screening. For the SampleStream platform component of this broader workflow, two key things were illustrated. Perhaps the more important of the two, is that co-elution of half antibodies along with full antibodies does not suffer from significant bias due to different ionization efficiencies, except when the half antibody makes up >50% of the sample (not a good clone anyway). A total of 192 samples from multiple platforms all demonstrated this same behavior. The second important aspect is running at 1 min/sample. While there are other platforms capable of this level of throughput, the removal of a chromatographic resin, and the non-specific interactions that come along with it, make the SampleStream platform less sensitive to variations in properties such as hydrophobicity or pI. Take a look at this manuscript by Benjamin Des Soye and colleagues at Northwestern University. Their research heavily leveraged the Plugin System from CTC Analytics AG, a tool that empowers programmers to teach the PAL 3 System how to control virtually any hardware with a digital communication interface. This allows the robot to seamlessly integrate with external hardware without the need to modify the drivers that link it to MS data acquisition software. Essentially, the additional hardware appears as native modules within the acquisition software. For instance, the SampleStream modules are entirely dependent on this system. In their work, the authors employed a plugin for the KingFisher, an automation platform designed for affinity purification, enabling immunoprecipitations (IPs) to be completed before processing samples via the SampleStream Platform—all within individual rows in the run list. IPT has also developed plugins for other devices, such as 2D barcode readers and heater/shakers. The flexibility of the plugin system gives IPT the ability to bring more hardware under automated control, creating comprehensive automation solutions. Here is a quick little "capability note". A sample of NISTmAb with a 3.5x excess of BSA was processed in a flow cell with a 10 kDa MWCO membrane then transferred to a flow cell with a 3 kDa MWCO membrane for reduction. The BSA was completely removed. Sacrificial proteins are sometimes used as "blocking" materials to improve recovery from vials. Additionally, excess Fc has been used for native elution from affinity capture. Subsequent processing of this sample type on the SampleStream Platform can easily remove these high abundance species prior to analysis.
