Messenger RNA (mRNA) vaccines are highly effective against viral infections, but researchers are seeking more stable and efficient alternatives for cancer immunotherapy. Circular RNA (circRNA), known for its stability and low immunogenicity, has emerged as a promising option. Researchers developed small circRNA vaccines in lipid nanoparticles, which induced strong and lasting T cell responses in mice. These vaccines showed higher efficacy, stability, and lower toxicity than traditional mRNA vaccines. When combined with immune checkpoint inhibitors, they effectively suppressed resistant tumors, highlighting their potential for cancer immunotherapy. Researchers from Shanghai Jiaotong University School of Medicine etc. published their research Delivery of FGF18 using mRNA-LNP protects the cartilage against degeneration via alleviating chondrocyte senescence on Journal of Nanobiotechnology. Osteoarthritis (OA) is a degenerative joint disease with significant unmet medical needs, and FGF18 protein has shown potential for cartilage repair, though its therapeutic efficacy is limited by poor delivery and short half-life. This study demonstrates that lipid nanoparticle (LNP)-delivered FGF18 mRNA can penetrate deeper into cartilage and sustain FGF18 expression for up to six days through mRNA UTR optimization and chemical modification. The treatment activates the FOXO3a-autophagy pathway, protecting chondrocytes from degeneration and senescence, and significantly alleviates OA symptoms in preclinical models. These findings highlight LNP-mRNA as a promising therapeutic strategy superior to recombinant FGF18 protein for OA treatment.
