The European Organ-on-Chip Society (EUROoCS) is an independent, not-for-profit organization

The mission of EUROoCS is enable Organ-on-Chip technology to facilitate life science research, promote human health and improve quality-of-life for all patients. To achieve this, EUROoCS aims to facilitate OoC scientists, industry, government regulators and the general public in realizing sustainable progress in biological and medical research through education, communication, funding and matchmaking. Beneficiaries include the pharmaceutical-, cosmetics-, chemical- and agri-food industries with impact on consumer products. EUROoCS will thus be the catalyst for change around the experimental use of animals. Through EUROoCS, OoC technology will become standardised and reproducible, promoting adoption by regulators, academia, industry and the general public, towards improving human health and safety.

?? This week we have another exciting new research to share from one of our members, Mara Lucchetti, who conducted a study on a novel gut-liver platform which provides new strategies for drug development and personalized medicine. ?? The human microbiome significantly influences drug metabolism through the gut–liver axis, leading to modified drug responses and potential toxicity. Understanding the microbiome-driven impacts on drug metabolism is limited due to the complex nature of the human gut environment. To address this challenge, Mara and her co-workers from the Systems Ecology group at the Université du Luxembourg, along with Alexander Mosig's lab, have developed a multiorgan-on-a-chip (MOoC) system, mimicking the bidirectional interconnection between the gut and the liver known as the gut-liver axis. In a proof-of-concept study, the metabolism of irinotecan, a widely used colorectal cancer drug, was simulated within the MOoC. The researchers tracked irinotecan metabolites using liquid chromatography coupled to tandem mass spectrometry, confirming the platform's ability to represent drug metabolism along the gut–liver axis. Additionally, the study demonstrated that the colorectal cancer-associated gut bacterium, Escherichia coli, modifies irinotecan metabolism by transforming its inactive metabolite SN-38G into its toxic metabolite SN-38. This finding highlights the platform's robustness in investigating the intricate interplay between gut microbes and pharmaceuticals. "Our gut–liver platform, validated with irinotecan, lays the foundation for studying other drugs affected by gut microbes," the researchers noted. The MOoC platform serves as a powerful tool for exploring the complexities of drug metabolism influenced by the gut microbiome, offering a representative alternative to animal models. "Future applications of our platform hold promise for expanding our understanding of drug metabolism and microbiome interactions, contributing to advancements in personalized medicine and therapeutic interventions," the team added. ??
