Human Cord Blood Exosomes Exosomes, tiny vesicles secreted by human umbilical cord blood mesenchymal stem cells (hUCB-MSCs), hold remarkable potential in various therapeutic applications. Recent research highlights their ability to stimulate regenerative wound healing by inhibiting the transforming growth factor-β receptor. Additionally, exosomes derived from hUCB-MSCs have shown promise in alleviating Parkinson's disease and neuronal damage through microglia inhibition. Moreover, studies suggest that umbilical cord blood plasma-derived exosomes possess antifibrotic effects, offering potential as a cell-free therapy for liver fibrosis. These exosomes also exhibit rejuvenating properties for human skin and accelerate cutaneous wound healing by promoting fibroblast function, angiogenesis, and M2 macrophage differentiation. Furthermore, they modulate interleukin-2 signaling in T cells, providing a potential therapeutic avenue for autoimmune diseases like experimental autoimmune encephalomyelitis. Notably, exosomes secreted by hUCB-MSCs attenuate diabetes in mice and enhance angiogenesis through the Wnt4/β-Catenin pathway, further underscoring their versatility and therapeutic potential. Their ability to autonomously navigate toward the liver and cross the blood-brain barrier stems from their interaction with transferrin and its receptors. Utilizing raw plasma exosomes as carriers for delivering drugs to the brain is a feasible and promising approach. Compared to cellular therapies, exosomes present a viable alternative, as they can efficiently transport desired molecules without the inherent limitations of cell-based treatments. Their easy accessibility, safety, and versatility make them ideal vehicles for therapeutic agents, including biomolecules and nucleic acids. Bovine Milk Exosomes Milk-derived exosomes are gaining attention as bioactive components of breast milk, demonstrating resilience against gastric digestion and reaching systemic circulation, thereby becoming bioavailable upon oral consumption. These exosomes harbor a diverse array of bioactive cargos, exerting beneficial effects across various domains, including immunity, intestinal and musculoskeletal health, gut microbiota, growth, and development. Encapsulated cargos within exosome membranes are shielded from enzymatic and non-enzymatic degradation, ensuring their integrity and functionality. However, the pasteurization process can diminish exosome concentrations by approximately 50%, attributed to membrane disruption and cargo degradation. Nevertheless, milk exosomes serve as nature's abundant carriers for drug delivery, boasting superior stability, biocompatibility, extended half-life, and minimal immunogenicity. Engineered milk exosomes hold promise for advanced theranostic applications, particularly in targeted drug delivery. Bovine milk-derived exosomes have emerged as a viable nano-vehicle for encapsulating and delivering macromolecular biotherapeutics, owing to their negatively charged hydrophobic nature, enabling the penetration of multiple biological barriers for oral drug delivery. Purified milk-derived exosomes have demonstrated the capacity to be directly taken up by keratinocytes and fibroblasts in vitro, leading to the upregulation of natural factors associated with skin moisturization, such as filaggrin (FLG), aquaporin 3 (AQP3), and CD44 in keratinocytes, as well as hyaluronidase (HAS2) in fibroblasts.
