?? New preprint alert!! EpiCypher is excited to share the results of our collaboration with Deepak Jha, George Q. Daley, MD, PhD Mohamad Ali Najia Margaret Shipp, which identified novel epigenetic targets for diffuse large B-cell lymphoma (DLBCL) therapy. Specifically, this research showed that DLBCL is highly sensitive to KDM4A and KDM4C inhibitors. How does it work? Here is a quick breakdown: ? Inhibition of H3K9 demethylases KDM4A & KDM4C rewires heterochromatin in DLBCL ? Rewiring of heterochromatin induces DNA replication stress cell-intrinsic inflammation ? These changes are highly cytotoxic to DLBCL cells - pointing to an key therapeutic vulnerability ? Bonus! This work leveraged EpiCypher's dCypher? platform for high-throughput screening to characterize novel KDM4 inhibitors. Unlike other platforms, dCypher assays use nucleosomes, which accurately reflect chromatin enzyme activity. https://lnkd.in/ek6xxyn7

Fiber-seq is a method developed by the Stergachis lab, that allows users to map chromatin accessibility on long-read sequencing platforms. First published in 2020, Fiber-seq has been growing in popularity - especially with the recent publication by the Stergachis lab, leveraging Fiber-seq for multiomic interrogation of Mendelian conditions (https://lnkd.in/dyyPQ4xm). As noted in the Genome Web article, many labs have been starting to implement this tech - from stem cells, to comparative genomics research, and beyond. ?? So where does EpiCypher come in? Our goal is to standardize Fiber-seq and provide kits, enzymes, and services for the scientific community. We want to make this strategy more accessible to help advance epigenetics research!
