    Interesting post and C&EN reporting on endotoxin testing, which conventionally has relied on horseshoe crab blood to produce a high-sensitivity reagent called limulus amoebocyte lysate (LAL). New animal-free recombinant tests have been approved by the US Pharmacopeia and are expected to go into effect in May 2025. Using Plasmid DNA as a template for IVT-mediated mRNA synthesis requires rigorous endotoxin testing to ensure that contaminants from bacteria used in the cloning process are removed before downstream use. In contrast, ELEGEN’s next-gen #genesynthesis is cell-free, completely eliminating cloning and the risk of contaminants such as endotoxins, mycoplasma, and residual genomic DNA to enable an efficient and more environmentally friendly workflow.     No crabs were harmed in the making of this post; but millions have died for the sake of biotech. The endotoxin testing market is worth about $1B annually in the US alone. Any substance or medical device going into a patient or animal has to be tested for endotoxin to ensure there are no trace amounts of glycolipid from the surface of bacteria. Unfortunately for the horseshoe crab their blue colored blood contains a sensitive biosensor that enables simple detection of endotoxin. This was first discovered in 1964 and the first endotoxin tests using LAL from horseshoe crabs was approved by the FDA in 1977. Fast forward to today and almost a million crabs are collected in the US each year. They are bled and then released but up to 30% later die and those that survive may not be healthy enough to spawn. It's difficult to imagine the biotech industry without these simple and accurate endotoxin tests. For decades it has been a necessary evil. But it might surprise you to learn that an animal free recombinant version of the critical protein was developed as far back as 1995. This was then commercialized by Lonza in 2003 but still to this day recombinant based endotoxin tests haven’t really taken off. In the EU these recombinant tests were officially recognized in 2015 and in China this occurred in 2020 but the US has lagged behind. The small number of producers of horseshoe crab based tests dominate the market and potentially have a lot to lose with a wholesale switch to recombinant. There have been many false dawns in the fight to save the horse shoe crabs from unnecessary harm but finally we might be at a real turning point. In November, US regulators will formally announce the acceptance of recombinant alternatives for endotoxin testing, which should then come into effect in May 2025.     IVT mRNA technologies are showing great promise as next-gen therapeutics, though there remain questions about the effect of impurities that can occur during the #IVT reactions re: the functionality and stability of the mRNA. Great step forward by Julien Camperi, Axel Guilbaud and the Genentech team on developing highly sensitive and advanced methods to characterize these IVT mRNA impurities to ensure quality control of RNA-based therapies! One method studied PolyA tail length on IVT, showing a positive correlation of IVT efficiency between longer tails (160 nt) and protein expression.
