Human umbilical vein endothelial cells (HUVECs) are a preferred #invitro model of endothelial cells, used to study vascular processes like angiogenesis, inflammation, and tissue repair. HUVECs are also used to study the integrity of the endothelial tissue barrier, which regulates the exchange of biomaterials between the bloodstream and surrounding tissues. Examining how these cells proliferate and from barriers is critical to understanding pathological and developmental processes in #vascularbiology. In this case study, we use Pixel electrical imaging to monitor in vitro HUVEC behavior in real time, including responses to barrier modulations. Sensitive detection of cell death in physiologically relevant in vitro models is critical to improving CAR-T cell therapies. With single-cell resolution, Pixel electrical imaging allows you to closely monitor interactions between T and tumor cells, including changes in cell morphology before death. Here, dying cells with altered morphologies are visible in green and blue. In this experiment, HeLa-CD19 cells were incubated with CD19-targeting T cells from ProMab Biotechnologies, Inc. for 72 hours. The left panel shows untreated cells, while the middle and right panels are seeded with 1:6 and 1:10 CAR-T:tumor cells at 24 hours, respectively. Cell death is visible even at low CAR-T:tumor cell ratios. #CARTcells #immunooncology #TcellTherapy

Endothelial cells form a selectively permeable barrier in vascular tissues that regulates the exchange of materials like ions, nutrients, and immune cells between blood vessels and the surrounding environment. Altering the permeability or structural integrity of this barrier can lead to conditions like inflammation and cardiovascular disease. How do CAR-T cells attack tumors in real-time? Pixel electrical imaging captures the dynamic interactions between T cells and tumor spheroids, revealing both structural changes and immune cell activity. In this experiment, HeLa-CD19 spheroids were incubated with CD19-targeting T cells from ProMab Biotechnologies, Inc. for 72 hours. The left panel shows an untreated spheroid growing over time, while the right demonstrates CAR-T cells reducing the tumor mass within 24 hours after their addition. Pixel enables visualization of T cell movement as well as their impact on the tumor microenvironment. #CARTcells #immunooncology #TcellTherapy

Primary endothelial cells are critical to modeling vascular pathologies in a variety of diseases, such as cardiovascular disorders and metastatic cancers. Capturing variations in these cell populations across individual donors is crucial to predicting efficacy of targeted therapies. In this case study, Pixel electrical imaging was used to monitor functional differences in human coronary artery endothelial cells (HCAECs) with varying donor and cellular characteristics. The resulting multiparametric data provide key insights for enhancing the clinical relevance of in vitro cellular assays and models.
