This study reveals how IRE1α interacts with stress granules (SGs) to strengthen cellular resilience during ER stress. Through ER membrane-bound phase separation, IRE1α forms clusters that tether to SGs via intrinsically disordered regions, amplifying the pro-survival IRE1α–XBP1 pathway for efficient UPR activation. Disruption of SG assembly impairs IRE1α clustering and reduces stress-handling capacity, underscoring the vital role of IRE1α–SG coalescence in preserving cellular homeostasis. ??
