Over the last decade, #ADCs have emerged as a significant class of targeted cancer therapies. In this webinar, Dr. Fu Li, vice president of biology at Pheon Therapeutics, will delve into the critical components of ADC design and how advancements in each area have led to the development of increasingly sophisticated ADCs now in clinical trials. This new paper was published in Nature Cancer and is entitled “An antibody–toxin conjugate targeting CD47 linked to the bacterial toxin listeriolysin O for cancer immunotherapy”. The authors from The University of Texas MD Anderson Cancer Center?engineered an antibody–toxin conjugate (ATC) that targets the ‘don’t eat me’ signal CD47 linked to the bacterial toxin listeriolysin O from the intracellular bacterium Listeria monocytogenes via a cleavable linker (CD47–LLO). CD47–LLO treatment in vivo significantly inhibited the growth of both localized and metastatic breast and melanoma tumors and improved animal survival as a monotherapy or in combination with checkpoint blockade. ATC may be regarded as a special ADC. Sanofi has entered an deal to buy Dren Bio, Inc.’s bispecific antibody DR-0201 for $600 million upfront payment, plus up to $1.3 billion in certain development and launch milestone payments. Congratulations! DR-0201 is a first-in-class bispecific antibody capable of engaging tissue-resident and trafficking myeloid cells to induce deep B cell depletion via targeted phagocytosis. This new paper looks interesting to us. The title of this open access paper is “Deep learning-based design and experimental validation of a medicine-like human antibody library”. The authors describe a deep learning model for computationally generating libraries of highly human antibody variable regions whose intrinsic physicochemical properties resemble those of the variable regions of the marketed antibody-based biotherapeutics (medicine-likeness).
