Our Founder, Dr. Anne Chiaramello is eager co-present at the first Apéro des Sciences of the season, with Dr. Nicolas Bouladoux, Staff Scientist, Laboratory of Host Immunity and Microbiome, National Institute of Allergy and Infectious Diseases (NIAID), NIH. What is Apéro des Sciences? You might say these events encourage scientific minds to unwind and enjoy the French joie de vivre (joy of living:) Apéro is a beloved evening tradition enjoyed in French culture. Short for apéritif, this word has taken on a meaning far beyond the drink itself. The congenial and relaxed apéro experience is a classic representation of the French. Juxtaposed with scientific insights, Apéro des Sciences is a series of events that engage the French and Francophile scientific community. The events generate thought-sharing and build a familial, cross-culture community amongst scientific minds. Chiaramello, who completed her first two degrees in her homeland, will share highlights of what led her to set sail for San Diego four decades ago, to complete her education, followed by her early-career experiences at Colorado State University. After the Apéro Evening, Dr. Chiaramello will unpack those intriguing highlights of her journey to our followers HERE on LI (in apéro-style tidbits:) Of course, our tenacious Founder will give insights into the latest progress the Mito-EpiGen Program’s groundbreaking research! Chiaramello gives a sneak peek, explaining, “We are at a momentous point in our FDA-authorized Basket Clinical Trial.” She smiles, “That’s all that I will share until after l’apéro.” So, stay tuned! A thought piece about why this is such a good time to invest in rare disease therapeutics.

The Chiaramello Lab and Mito-EpiGen Program affectionately calls the infographic below our “Wavy Arrow”.?With so much packed in, one can easily overlook the full gravity of our journey; we are now sharing the reality of choosing the path of academic integrity and research autonomy from Big Pharma funding. Last month, we celebrated the long-awaited shipment of customized, enteric Capsugel Enprotect capsules, from France. The Lab now proudly shares their arrival at the clinical drug manufacturer PMRS, Inc., in Philadelphia, PA. The?active pharmacological ingredient (investigational drug) manufactured?in the US by Penta Manufacturing Company, in Livingston, NJ also arrived at PMRS; clinical drug manufacturing with this innovative drug delivery system is underway! In terms of the infographic, we're at Step-4. Prior to this momentous milestone, the Lab Team invested 10+ years harvesting patient-derived cells and deciphering each patient's mitochondrial signature in keeping with applying Personalized Medicine instead of one-size-fits all therapeutic model. Steps 1-2 included mapping mitochondrial pathogenic variants.?This led to breakthrough translational research, revealing mitochondrial signatures shared by two rare diseases, LHON-Plus & MELAS. This discovery resulted in a National Institute of Neurological Disorders and Stroke (NINDS) translation grant (for MELAS), and United States Department of Defense discovery award (for LHON-Plus).? In Step-3, National Center for Advancing Translational Sciences (NCATS) then provided funding to pivot from translation to clinical research; The Lab’s Mito-EpiGen Program received approval for the first-ever FDA-authorized Basket Clinical Trial for rare mitochondrial diseases.?With such small patient populations, combining two rare diseases is critical to a successful trial.?? Dr. Anne Chiaramello shares, “We took one leap of faith after another. Immeasurable hours were invested just to get to Step-1 in our Wavy Arrow. It then took years of experimentation/tinkering with collaborative and innovative partners, Capsugel & Lonza, to create the precisely right drug delivery system to optimize patient-specific dose of the investigational drug. We assembled and synchronized global collaborators and components for US-based clinical manufacturing to begin, with regular FDA inspection. THOSE LEAPS OF FAITH ARE COMING TO FRUITION. While it is gratifying, we must focus on getting through & beyond Step-4, by bridging the funding gap.” Patient Advocate, Andrea Hoffmeier further explains, “Why the funding gaps??The Lab made a deliberate decision of integrity in patient-focus (not profit-influenced). The process to qualify every potential patient costs thousands; when one is disqualified, it's a lost investment. Clinical drug manufacturing is also costly; each capsule pill more like hand-tailoring, than fast-fashion factory apparel. We WILL bridge the gap, with a multi-pronged strategy, depicted in the Wavy Arrow. Stay tuned!”

Amazing Training Opportunites with the National Center for Advancing Translational Sciences (NCATS) Dr. Anne Chiaramello shared her viewpoint of the recent post https://lnkd.in/gHWCqZEe “NCATS offers a unique opportunity for young investigators to launch their career in translational science by training in the NCATS innovatory labs.” Chiaramello encourages Young Science Scholars, Fellow Educators and Science-Focused Mentors to check out the the NCATS Page details: https://lnkd.in/g4eDscaE We offer novel and holistic training opportunities for early career scientists, from undergrads through postdocs. Trainees gain translational science skills through hands-on research and a range of educational and professional development opportunities. Chiaramello explains that the interdisciplinary nature of the NCATS Training Program is especially important. “The tapestry image pictured below resonates with me now, more than ever. To achieve our goals in our MitoEpi-Gen Program at The George Washington University School of Medicine and Health Sciences, we cannot operate in a silo. The Lab collaborates with other researchers, suppliers, including Capsugel, PMRS, Inc. and Lonza, as well as our clinical partner, Children's National Hospital. NCATS DPI research teams encourage interdisciplinary interactions, and it is inspiring to see them organized into four interwoven branches.” The NCATS webpage explains, “Many of these research teams use similar technologies, skills and expertise …resulting in substantial collaboration among disciplines as depicted by the overlapping ribbons [see image below]. The complementarity of expertise …creates an enriching environment in which fellows learn multiple skills from domain experts and gain additional scientific and career mentorship.” This program is a vital element of the NCATS 5-Year Strategic Plan, as shared by Dr. Joni L Rutter. Many in my generation of scientists might regret being too advanced in their career to participate in this program. So, do the next best thing - encourage an up-and-coming scientist to grab hold of this amazing opportunity! The celebration of the 2024 Nobel Prize recipients in Physiology or Medicine is the perfect timing for highlighting the Nobel Prize time machine of The George Washington University School of Medicine and Health Sciences by showcasing the groundbreaking discovery of Dr. Julius Axelrod, a #GWSMHS graduate, on the release and reuptake of catecholamine neurotransmitters. In 1955, Dr. Julius Axelrod was awarded the Nobel Prize in Physiology or Medicine for his discovery that laid the foundation for the development of many anti-depressants targeting the reuptake of neurotransmitters. What a wonderful time to honor Dr. Julius Axelrod during the #GWSMHS bicentennial celebration! From l’apéro (a joyful evening gathering), to a momentous air shipment of a customized drug delivery system, this is and exciting time for not just ONE, but TWO Collaboration Announcements: #1-APéRO DES SCIENCES It was a great honor and privilege for our Founder, Dr. Anne Chiaramello to showcase her global academic path from basic research to clinical research funded by the National Center for Advancing Translational Sciences (NCATS) before the team of Mireille Guyader PhD, Counselor for Science and Technology for the Embassy of France in the United States. This was the first Apéro des Sciences to be hosted outside the walls of the French Embassy, at The George Washington University! Alison K. Hall, PhD, Senior Associate Dean of Research at the The George Washington University School of Medicine and Health Sciences, shared her thoughts on the importance of our founder’s tenacity. “Anne has a knack to elevate and convert basic research into clinical opportunity that has led to the novel concept of basket clinical trial for rare mitochondrial diseases. She is the only one at GW SMHS who made it happen. This is why we are delighted and honored that Dr. Mireille Guyader invited Dr. Chiaramello to the first Apéro des Sciences held in the Science and Engineering building of The George Washington University.” #2-PRECIOUS CARGO - FRANCE-USA Concurrently, we celebrate the long-awaited shipment of the customized and enteric Capsugel Enprotect capsules have left France and are headed to the USA clinical drug manufacturer PMRS, Inc., located in Philadelphia, PA! This is a HUGE milestone. It took years of experimentation and tinkering with a truly collaborative and innovative partners, Capsugel in France and Lonza in the US, to create the precisely right drug delivery system to optimize patient-specific dose of the investigational drug. The pro-drug itself is produced in another state, New Jersey, so the triangle of components and The Lab have all come together for clinical manufacturing to begin. ? SO THE DREAM IS BECOMING REALITY! ? The actual clinical manufacturing process requires a highly specialized state-of-the-art cGMP facility. The team at PMRS has the expertise to make it happen with high industry standards congruent with FDA regulations.
