It’s been an incredible week in Sydney, Australia where I had the opportunity to give a talk on my research during the Tumor Immunology session at the 52nd Annual Meeting for the Australian and New Zealand Society for Immunology (ASI)! A major hurdle in developing TCR-T cell-based immunotherapies for the treatment of cancer is our insufficient knowledge of what drives immunogenicity. The goal of my research project is to comprehensively assess the factors contributing to differences in tumor immunogenicity across different classes of antigens and define the principles of CD8+ T cell immune recognition that shape these responses. Interestingly, we found that the number and diversity of the neoantigen-specific responses varied between both the types of antigens and donors tested. I’m excited to continue leading this research to help develop improved cell-based immunotherapies for patients with cancer. I would like to thank Drs. Pick any paper. If it involves the protein, nucleic acid, secreted content or behavior of prokaryotic or eukaryotic cells, there's likely a subsection describing how those cells were grown in culture - whether in petri dishes, multi-well dishes, flasks or 96-well plates. Cell culture is how researchers expand cell clones and colonies to harvest macromolecules, enrich specific phenotypes or to interrogate their responses to changing biological conditions or chemical treatment. Integral to such work is the unquestionable assumption that all the cells in a dish will be identical— so by growing and interrogating them in culture, the researcher is simply isolating and amplifying the signal. Right? - But is this always true? - Might subtle differences at genotypic and even phenotypic levels yield significant variation in cellular behavior which may be averaged out across the population? - Could these differences have significant downstream impacts long-term on the stability of your master cell bank, binding affinity of your biologic or either rapid exhaustion or cytotoxic cytokine profile of your activated T cell? Until recently researchers had no way to probe this variability. Today, they do: The Beacon Optofluidic System - with single cell cloning and constant perfusion - provides targeted, accurate and sensitive profiling of single cell identity and behavior for 1000s of cells over the life-time of each cell.
