New #publication spotlight from PLOS Pathogens using fluorescent imaging from the Sapphire Biomolecular Imager to determine IC50 using in-cell Western. ?? While the emergency phase of the pandemic is considered to be over by the WHO, COVID-19 remains a circulating respiratory virus that continues to evolve and cause illness globally. In particular, SARS-CoV-2 continues to mutate, leading to new subvariants with varying degrees of transmissibility and immune evasion. In a PLOS Pathogens paper, Dong et al isolated several promising candidates for disrupting the cycle of immune selection and viral escape through effectively blocking respiratory acquisition and transmission of diverse sarbecoviruses. The Sapphire Biomolecular Imager was used to determine the neutralizing activity of the top nanobody candidates through fluorescence in-cell Western. Obtaining high-quality purified proteins is essential for structural biology experiments. Membrane protein purification is particularly complex, as evidenced by the fact that less than 1% of entries in the Protein Data Bank represent membrane proteins. A method put forth by Drew et al. (2008) involving SDS-PAGE has proven useful in the purification of fluorescently labeled membrane proteins. Our recent app note explores an adapted protocol, in which the Sapphire Biomolecular Imager provides optimized in-gel fluorescence detection for semi-quantifying and monitoring the purification of GFP-labeled membrane proteins from yeast.
