Attune Neurosciences, INC. is conducting a research study in San Francisco, California to evaluate a non-surgical, non-drug wearable therapy using Low Intensity Focused Ultrasound Stimulation (LIFUS) of the deep brain to alleviate pain. Conducting this research study will enable further development of a novel wearable device, to allow in-clinic or at-home use for pain reduction. This technology is already being used in multiple studies for other conditions. LIFUS is a form of non-invasive brain stimulation that uses sound waves (the same form of energy used in fetal ultrasound imaging during pregnancy) to target and alter activity in areas of the deep brain. The massive symptom variability across depression patients suggests that the brain areas most in need of treatment may also be different. At Attune Neurosciences, Inc., we have developed a wearable, offline, MR-guided ultrasound neuromodulation device that can be comfortably worn for hours, even overnight. This allows us to non-invasively explore personalized treatment solutions for every patient. In collaboration with leading UCSF researchers, we iteratively examined the efficacy of numerous brain areas involved in treatment resistant major depressive disorder. While we have lots of exciting data to come, we wanted to share the experience with our first patient, who reliably experienced symptomatic relief from stimulation of the Anterior Thalamus. Notably, structures just a few millimeters away showed no benefit, underscoring the remarkable spatial specificity researchers can achieve with focused ultrasound. Surprisingly, ANT stimulation reduced the patient's default mode network connectivity from the 99th percentile of over-connectivity back to healthy levels—a finding with massive implications.
